荧光激活蛋白研究进展

摘要 荧光蛋白历经三十余年的快速发展,已经成为生命科学领域不可或缺的研究工具,在生物学、医学和生物工程等领域有着十分广泛的应用.与有机荧光染料相比,荧光蛋白存在的亮度低、光稳定性差和光谱可调性差等不足限制了其在活体标记、超分辨成像等方面的应用.荧光激活蛋白是一类可特异性结合小分子荧光配体,并显著激活配体产生荧光的蛋白质标签.相较于传统的荧光蛋白,荧光激活蛋白不仅具有可遗传编码的特点,同时还具有小分子荧光配体的亮度高、光稳定性好及光谱可调的优点.近十年来,研究者基于不同的策略,设计构建了一系列荧光激活蛋白,这些荧光激活蛋白已经在超分辨成像、分子传感等领域得到了广泛应用.荧光激活蛋白及其衍生技术的发展源于研究者对生命和化学规律的深入理解,相关技术的发展将推动新一代生物技术的快速发展. Over the past thirty years,fluorescent proteins have become indispensable tools in the field of life science,with widespread application in biology,medicine and bioengineering.However,compared to organic fluorescent dyes,fluorescent proteins exhibit limitations such as low brightness,poor photostability,and limited spectral tunability.These drawbacks hinder their use in vivo labeling and super-resolution imaging.Fluorescence-activating proteins represent a class of protein tags that can specifically bind to non-fluorescent small molecule fluorogenic ligands and significantly activate their fluorescence.Compared to traditional fluorescent proteins,fluorescence-activating proteins are not only genetically encodable,but also offer advantages such as high brightness,good photostability and high spectral tunability.During the past decade,researchers have designed and constructed various fluorescence-activating proteins using different strategies,and these proteins have been extensively utilized in super-resolution imaging,molecular sensing and other fields.The development of fluorescence-activating proteins and their derived technologies stems from researchers’deeper understanding of the principles of life and chemistry,which will propel the rapid development of a new generation of biotechnology.
