肿瘤相关成纤维细胞与化疗抵抗关系的研究进展

摘要 肿瘤相关成纤维细胞(cancer associated fibroblasts,CAFs)是肿瘤微环境中数量最多的基质细胞,具有高度异质性和来源广泛性,是肿瘤间质中各种细胞之间通信的枢纽,与化疗抵抗密切相关。有别于既往研究只关注肿瘤细胞自身,该文着眼于肿瘤微环境中的基质细胞CAFs,对其来源、异质性以及CAFs促化疗抵抗相关机制(释放外泌体、调节信号通路、调节自噬、促进癌细胞干性化、调控细胞衰老)进行梳理,同时对调控CAFs改善化疗抵抗策略进行探讨,旨在探索靶向CAFs应对化疗抵抗的新策略,给临床以启示。 Cancer associated fibroblasts(CAFs)are the most abundant stromal cells in the tumor microenvironment,with high heterogeneity and extensive sources.They are the communication hub between various cells in the tumor stroma and are closely related to chemotherapy resistance.Unlike previous studies that only focused on tumor cells themselves,this article focuses on stromal cell CAFs in the tumor microenvironment,and reviews their sources,heterogeneity,and mechanisms related to CAFs promoting chemotherapy resistance(releasing exosomes,regulating signaling pathways,regulating autophagy,promoting cancer cell stemnesis,and regulating cell aging).At the same time,it explores strategies for regulating CAFs to improve chemotherapy resistance,aiming to explore new strategies for targeting CAFs to respond to chemotherapy resistance and provide insights for clinical practice.
