前列消汤通过IL-6/JAK2/STAT3通路调控自身免疫性前列腺炎炎症反应的机制

Study on mechanism of Qianliexiao Decotion regulating the inflammatory response of experimental autoimmune prostatitis through IL-6/JAK2/STAT3 pathway

摘要 目的:探讨前列消汤通过白细胞介素6(IL-6)/Janus激活激酶2(JAK2)/信号转导激活因子3(STAT3)通路调控自身免疫性前列腺炎(EAP)炎症反应的机制。方法:将雄性SPF级C57BL/6小鼠180只按随机数表法分为空白组,模型组,阳性对照组(AG490抑制剂),前列消汤低、中、高剂量组;采用复合因素制备EAP湿热证小鼠模型,以前列腺湿重指数、HE评估模型,Western Blot、RT-PCR检测EAP小鼠前列腺组织IL-6、糖蛋白130(gp130)、JAK2、STAT3、信号转导抑制因子3(SOCS3)、视黄酸相关核孤儿受体γt(RORγt)、白细胞介素17A(IL-17A)、叉头样转录因子3(Foxp3)蛋白及mRNA表达水平。结果:模型组前列腺湿重指数较空白组升高(P<0.05);与空白组比较,模型组单核、炎症细胞浸润及腺泡细胞壁增厚、腺腔结构紊乱,各药物组不同程度改善模型小鼠前列腺的炎症浸润程度,前列消各剂量组干预效果以高剂量组最为显著;与EAP组比较,除前列消汤低剂量组RORγt蛋白外,前列消汤各剂量组显著下调IL-6、gp130、JAK2、STAT3、SOCS3、RORγt、IL-17A、Foxp3蛋白及mRNA表达(P<0.01,P<0.05)。结论:前列消汤通过IL-6/JAK2/STAT3通路抑制EAP炎症反应,进而改善EAP小鼠前列腺病理结构及自身免疫反应功能。 Objective: To study the mechanism of Qianliexiao Decoction regulating the inflammatory response of experimental autoimmune prostatitis(EAP) through IL-6/JAK2/STAT3 pathway. Methods: A total of 180 male SPF-grade C57BL/6 mice were randomly divided into blank group, model group, positive control group(AG490 inhibitor), low/medium/high dose Qianliexiao Decoction group. EAP mouse models with damp heat syndrome were prepared by using complex factors,and evaluated by prostate wet weight index and HE staining pathological model. Western Blot and RT-PCR were used to detect IL-6, gp130, JAK2, STAT3, SOCS3, RORγ t, IL-17A, Foxp3 protein and gene expression level in prostate tissues of EAP mice.Results: The prostate wet weight index in the model group was higher than that in the blank group(P<0.05). Compared with the blank group, the model group showed infiltration of monocytes and inflammatory cells, thickening of acinar cell wall, and disorder of glandular cavity structure. Each drug group could improve the degree of inflammatory infiltration of the prostate of model mice to varying degrees. The intervention effect of Qianliexiao Decoction in each dose group was most significant in the highdose group. Compared with the EAP group, except RORγt protein in low dose Qianliexiao Decoction group, the Qianliexiao Decoction groups down-regulated IL-6, gp130, JAK2, STAT3, SOCS3, RORγt, IL-17A, Foxp3 protein expression(P<0.01,P<0.05). Conclusion: Qianliexiao Decoction can inhibit the experimental autoimmune prostatitis reaction through IL-6/JAK2/STAT3 pathway, thereby improving the pathological structure and autoimmune reaction function of prostate in EAP mice.
