摘要
目的:采用肾脏代谢产物分析方法和肠道菌群评价蒙药冬葵果醇提物利尿作用机制。方法:50只SD大鼠随机分为正常组(生理盐水),冬葵果醇提物高、中、低剂量组(冬葵果醇提物1.6069、0.5356、0.1785 g/kg)和阳性对照组(氢氯噻嗪0.0214 g/kg)。采用代谢组学方法检测肾脏中内源性代谢产物,并进行代谢差异表征,查寻肾脏中差异代谢产物;采用16S rDNA测序技术检测冬葵果醇提物各剂量组对肠道菌群多样性及物种组成的影响,综合评价冬葵果醇提物利尿作用机制。结果:代谢组学分析显示,与正常组相比,冬葵果醇提物低剂量组肾脏样本中共筛选得出13个差异代谢物,冬葵果醇提物中剂量组28个差异谢物,冬葵果醇提物高剂量组14个差异代谢物,这些代谢物水平存在明显的变化。肠道菌群通过对各组OTU数据为计算依据绘制韦恩图,结果显示:与正常组比较,冬葵果醇提物各剂量组共有2146个OTU,其中冬葵果醇提物低剂量组独有416个OTU,冬葵果醇提物中剂量组独有381个OUT,冬葵果醇提物高剂量组独有330个OUT;Alpha多样性显示,冬葵果醇提物低、中剂量组肠道菌群丰度高于正常组,冬葵果醇提物高剂量组肠道菌群丰度低于正常组,初步认为冬葵果对正常大鼠肠道菌群有调节作用;门、属、科水平相对丰度差异比较,厚壁菌门、拟杆菌门、乳杆菌、S24_7、梭菌目、毛螺菌科、颤螺菌属、瘤胃球菌比重较高,在门水平上与正常组比较,冬葵果醇提物各剂量组的厚壁菌门相对增加,拟杆菌门相对减少;属和科水平上与正常组比较,乳杆菌在冬葵果醇提物低、高剂量组中减少;毛螺菌科在冬葵果醇提物各剂量组中增加。结论:冬葵果利尿作用机制可能与拟杆菌门-葡萄糖/葡萄糖酸、毛螺菌科-L-酪氨酸等两个菌群与内源性代谢产物通路有关。
Objective:To investigate the diuretic mechanism of the alcohol extract of Malva Fructus through the analysis of renal metabolites and gut microbiota.Methods:Fifty Sprague-Dawley rats were randomly divided into five groups:a normal group(saline),high,medium,and low dose groups of alcohol extract of Malva Fructus(1.6069,0.5356,0.1785 g/kg),and a positive control group(hydrochlorothiazide 0.0214 g/kg).Metabolomics was used to detect endogenous metabolites in the kidneys and characterize metabolic differences,identifying differential metabolites.16S rDNA sequencing was used to assess the diversity and composition of gut microbiota across different dosage groups of the alcohol extract,providing a comprehensive evaluation of its diuretic mechanism.Results:Metabolomics analysis showed that compared to the normal group,the low dose group of Malva Fructus alcohol extract had 13 differential metabolites,the medium dose group had 28,and the high dose group had 14,all showing significant changes in levels.The Venn diagram based on OTU data revealed that compared to the normal group,the Malva Fructus alcohol extract dosage groups shared 2,146 OTUs,with the low dose group having 416 unique OTUs,the medium dose group 381 unique OTUs,and the high dose group 330 unique OTUs.Alpha diversity indicated that the gut microbiota abundance in the low and medium dose groups was higher than in the normal group,while the high dose group had lower abundance,suggesting a regulatory effect of Malva Fructus on gut microbiota in normal rats.At the phylum,genus,and family levels,there were significant differences in relative abundance.Compared to the normal group,the Firmicutes increased and Bacteroidetes decreased across the Malva Fructus alcohol extract dosage groups.At the genus and family levels,Lactobacillus decreased in the low and high dose groups,while Ruminococcaceae increased across all dosage groups.Conclusion:The diuretic mechanism of Malva Fructus may be related to the pathways involving Bacteroidetes-glucose/gluconic acid and Rumin
