基于OmniLog TM系统评价鼠疫噬菌体在不同培养基中的生长情况

Evaluation of plague phage growth in different culture medium based on OmniLog TM system

摘要 目的2022年11月利用OmniLog TM微生物鉴定系统评价诊断用鼠疫噬菌体在赫金格尔消化液培养基(简称“赫氏肉汤”)、溶菌肉汤(luria-bertani,LB)液体培养基和IF-A培养基中的生长情况,为鼠疫噬菌体的筛选和生物学特性鉴定提供科学依据。方法以鼠疫弱毒菌EV 76株为宿主菌,基于OmniLog TM微生物鉴定系统,利用无菌96孔微量培养板在33℃环境作用48h,检测诊断用鼠疫噬菌体分别在赫氏肉汤、LB液体培养基和IF-A培养基中的生长情况。结果以赫氏肉汤、LB液体培养基为培养介质,试验列生长曲线几乎呈一条横线,且细菌浓度的对数值不超过100,对照组列中宿主菌呈对应的S型生长曲线;试验孔中四唑类染料的颜色随着噬菌体数量的减少和宿主菌数量的增多而逐渐加深。以IF-A培养基为培养介质,其试验列未显示任何生长曲线。结论三种液体培养基鼠疫噬菌体生长情况相比较,赫氏肉汤更优用于鼠疫耶尔森菌与噬菌体的相互作用研究,IF-A培养基更适合于其他微生物的生长。 Objective To evaluate the growth of plague phage for diagnosis in Hottinger digest liquid culture medium(Hottinger's broth for short),luria-bertani(LB)liquid culture medium,and IF-A culture medium by using the OmniLog TM microbial characterization system in November 2022,and to provide a scientific basis for the screening and biological identification of plague phage.Methods Using attenuated plague bacteria of EV 76 strain as the host bacteria,the growth of plague phage for diagnosis in Hottinger's broth,LB liquid culture medium and IF-A culture medium,respectively,was detected based on the OmniLog TM microbial identification system using sterile 96-well microculture plate operating at 33℃for 48h.Results Using Hottinger's broth and LB liquid culture medium as culture media,the growth curves of the test columns showed almost a horizontal line and the logarithmic value of bacterial concentration did not exceed 100,and the host bacteria in the control column showed a corresponding S-shaped growth curve.The color of the tetraazole dye in the test wells gradually deepened with the decrease in the number of phages and the increase in the number of host bacteria.The test columns did not show any growth curve using IF-A culture medium as culture culture medium.Conclusion Comparison of the growth of plague phage in the three liquid media,Hottinger's broth is preferable for the study on the interaction between Yersinia pestis and phage,and IF-A culture medium is more suitable for the growth of other microorganisms.
