摘要
目的基于国家知识产权局专利数据库中药复方专利,探析中药治疗糖尿病周围神经病变(DPN)的组方规律。方法收集建库至2023年12月符合纳入标准的中药治疗DPN的复方专利数据,运用R语言(v 4.3.2)、Cytoscape(Version 3.10.1)等软件对数据进行统计分析,采用数据建模、用药频次分析、性味归经分析、聚类分析、关联规则分析和数据可视化等以探讨DPN的组方规律。结果本研究共纳入符合标准的DPN专利数据113项,涉及中药249味,药物累计频次1145次。用药频次排名前10位的中药分别为黄芪、当归、丹参、川芎、桂枝、鸡血藤、赤芍、白芍、地黄、红花。中药药性以温、平、寒为主,药味以甘、苦、辛常见,多归肝、肾经。聚类分析共得到2个药物组合,关联规则分析共筛选出4组二项常用药对和32组三项关联药物组合,常用药对组合有鸡血藤-黄芪、延胡索-黄芪、桂枝-黄芪、桃仁-黄芪,常用角药组合有黄芪-延胡索-鸡血藤、当归-牛膝-川芎。中药共现网络显示,重要节点有黄芪、当归、丹参、桂枝、川芎、鸡血藤、赤芍等。结论治疗DPN的中药复方专利多用黄芪配伍当归、丹参、川芎、鸡血藤、赤芍、红花、川牛膝等活血药以益气活血通络,且多使用藤类药、虫类药通络祛邪,可为临床诊治DPN扩展思路。
Objective To explore the compound prescription rule of traditional Chinese medicine in the treatment of diabetic peripheral neuropathy(DPN)based on the patent database of the National Intellectual Property Office.Methods The compound prescription patent data of traditional Chinese medicine for DPN that met the inclusion criteria were collected from the inception to December 2023.The software of R language(v 4.3.2),Cytoscape(Version 3.10.1),and other software were applied to carry out statistical analysis on the data.Data modeling,drug frequency analysis,nature and flavour,channel tropism analysis,cluster analysis,association rule analysis,and data visualization were used to explore the formula composition rule of DPN.Results A total of 113 DPN patent data conforming to the standard were included in this study,involving 249 kinds of traditional Chinese medicines,and the cumulative frequency of drugs was 1145 times.The top ten most frequently used Chinese medicines were Astragali Radix,Angelicae Sinensis Radix,Salviae Miltiorrhizae Radix Et Rhizoma,Chuanxiong Rhizoma,Cinnamomi Ramulus,Spatholobi Caulis,Paeoniae Radix Rubra,Paeoniae Radix Alba,Rehmanniae Radix,and Carthami Flos.The drug natures of traditional Chinese medicine were mainly warm,plain,and cold,and the drug flavours were common to sweet,bitter,and pungent,which mostly return to the liver and kidney channels.A total of two drug combinations were obtained by cluster analysis,and four groups of binomial common couplet medicines and 32 groups of three associated drug combinations were screened by association rule analysis.The commonly used couplet medicines were Spatholobi Caulis-Astragali Radix,Corydalis Rhizoma-Astragali Radix,Cinnamomi Ramulus-Astragali Radix,and Persicae Semen-Astragali Radix. Commonly used horn medicine combinations were Astragali Radix-Corydalis Rhizoma-Spatholobi Caulis and Angelicae Sinensis Radix-Achyranthis Bidentatae Radix-Chuanxiong Rhizoma.
