摘要
为研究VD_(3)对高糖饲料喂养下黄鳝存活率及糖代谢的影响,实验选取健康的黄鳝幼鱼[初始体重(17.13±0.08)g]为实验对象,随机分为3组,每组4个重复,每个重复60尾鱼,分别投喂正常糖水平、高糖水平和高糖水平添加2000 IU/kg维生素D_(3)(分别命名CON组、HC组和HCVD3组)的3种等氮等脂饲料,进行为期8周的养殖实验。结果显示,各实验组生长性能及体成分均无显著影响;HC组存活率(82.22%)显著下降,添加VD_(3)后,存活率(88.89%)显著增提高,且与CON组(90.42%)无显著差异。HCVD3组脏体指数和肥满度较HC组均显著上升。血清谷丙转氨酶(ALT)和谷草转氨酶(AST)含量在HC组达到最大值;血清甘油三酯(TG)含量在HCVD_(3)组达到最大值。肠道淀粉酶和胰蛋白酶的活性HC组显著低于其他各组,而胰蛋白酶活性显著高于其他各组。HC组黄鳝肝脏中糖原含量(124.45 mg/g)较CON组(99.55 mg/g)显著升高,己糖激酶(HK)和磷酸果糖激酶(PFK)活性也显著升高;HCVD3组肝糖原含量(89.44 mg/g)较HC组显著下降,且PFK活性显著高于HC组。肌肉PFK活性在HCVD3组达到最高值。HC组黄鳝肝脏和肠道过氧化氢酶(CAT)活性均显著低于CON组,添加VD3后,总超氧化物歧化酶(T-SOD)均显著升高,丙二醛(MDA)活性均显著降低。HCVD3组黄鳝肠道促炎细胞因子(IL-1β、MyD88)、抗炎细胞因子(IL-10)和肠道紧密连接蛋白基因(ZO-1、ZO-2、Claudin-12)相对表达量显著优于HC组。研究表明,在本实验条件下,高糖(35%)日粮会显著降低实验鱼的存活率、增加肝糖原积累、降低其抗氧化能力并破坏肠道健康;而高糖饲料中添加2000 IU/kg VD_(3)后,可通过增强鱼体抗氧化能力和免疫力改善肠道屏障功能,进而提高鱼体存活率;此外,高糖饲料中添加VD_(3)会增强鱼体对糖的利用能力。本研究可为黄鳝的糖代谢调控和人工饲料的配制提供基础理论依据和指导。
To investigate the impact of dietary vitamin D_(3)(VD_(3))on the survival rate and glucose metabolism abil-ity of Monopterus albus fed high-carbohydrate diets,healthy M.albus juveniles(17.13±0.08)g were randomly assigned into 3 groups,each with 4 replicates of 60 fish.The fish were fed diets with normal,high carbohydrate level,and high carbohydrate level supplementing with 2000 IU/kg VD_(3)(referred to as the CON,HC and HCVD_(3) groups,respectively).After 8 weeks,no significant differences in growth performance or body composition were observed among the groups.However,The survival rate of HC group(82.22%)was significantly decreased,but after addition of VD_(3),SR(88.89%)was significantly increased,and the difference was not significant compared with CON group(90.42%).The visceral somatic index(VSI)and condition factor(CF)were significantly higher in the HCVD_(3) group than in the HC group.The HC group showed the highest levels of serum alanine aminotrans-ferase(ALT)and aspartate aminotransferase(AST),while the HCVD_(3) group had the highest serum triglyceride(TG)content.Intestinal amylase and trypsin activities were lower than in the HC group but higher in lipase.Liver glycogen content in HC group(124.45 mg/g)was significantly higher than CON group(99.55 mg/g),as were the activities of hexokinase(HK)and phosphofructokinase(PFK).In contrast,the HCVD_(3) group(89.44 mg/g)had lower liver glycogen but higher PFK activity.Muscle PFK activity peaked in the HCVD_(3) group.Additionally,catalase(CAT)activities in the liver and intestinal tract of M.albus were lower in the HC group,while total super-oxide dismutase(T-SOD)was increased,and malondialdehyde(MDA)activities were decreased with VD_(3) supple-mentation.The HCVD_(3) group also showed significantly higher relative expression levels of intestinal proinflam-matory cytokines(IL-1β,MyD88),anti-inflammatory cytokines(IL-10)and intestinal compact linking protein genes(ZO-1,ZO-2,Claudin-12)than the HC group.In conclusion,a high carbohydrate diet reduced the survival ra
