摘要
红花在临床上对急性酒精性肝损伤(Acute Alcoholic Liver Injury,AALI)有很好的疗效,但治疗机制不甚明确。因此,阐明红花治疗AALI的分子作用机制对药物的进一步开发及应用具有十分重要的意义。实验以雄性Wistar大鼠为研究对象,模型组大鼠灌胃8 mL/kg的酒精以建立AALI模型,给药组大鼠以4.29 g/kg/天的剂量灌胃红花提取物。利用大鼠血清代谢组学分析方法鉴定与AALI相关的潜在生物标志物,并研究红花对这些生物标志物的调控机制。从苏木精和伊红(H&E)染色实验发现,灌胃后红花提取物减轻了肝细胞的损伤程度;与模型组相比,红花组中的丙氨酸氨基转移酶(ALT)和天冬氨酸氨基转移酶(AST)的表达水平降低,表明红花提取物具有肝保护作用。通过分析火山图,共鉴定出30种与AALI相关的生物标志物。在这些生物标志物中,红花可以对21个标志物产生回调效果。研究发现,红花可能通过对脂肪酸、磷脂、胆汁酸、氨基酸、核苷酸代谢的调控作用从而对AALI产生治疗效果。本文为红花的推广和临床应用提供了理论基础。
Flos Carthami(FC)has a good therapeutic effect on acute alcoholic liver injury(AALI)in clinical practice,but the treatment mechanism is not very clear.Therefore,elucidating the molecular mechanism of action of FC in treating AALI is of great significance for the further development and application of drugs.The experiment focuses on male Wistar rats as the research object.The model group rats were gavaged with 8 mL/kg alcohol to establish an AALI model,while the administration group rats were gavaged with FC extract at a dose of 4.29 g/kg/day.Potential biomarkers related to AALI were identified using rat serum metabonomics analysis methods,and the regulatory mechanism of FC on these biomarkers was investigated.The H&E staining experiment found that after gavage,FC extract reduced the degree of liver cell damage;Compared with the model group,the expression levels of ALT and AST in the FC group decreased,indicating that FC extract has liver protective effects.Through volcanic map analysis,a total of thirty biomarkers related to AALI were identified.Among these biomarkers,FC can have a callback effect on 21 biomarkers.Research has found that FC may exert therapeutic effects on AALI by regulating the metabolism of fatty acids,phospholipids,bile acids,amino acids,and nucleotides.This article provides a theoretical foundation for the promotion and clinical application of FC.
