摘要
布尼亚病毒目包含超五百多种病毒,其中包含能引起严重人类和哺乳动物疾病的病毒,致死率高,传染性强,严重威胁人类健康和经济稳定。大多数布尼亚病毒的入侵机制尚不清晰,缺乏针对性的药物和疫苗。而受体研究是解析病毒侵染机制和探究药物靶点的关键步骤之一。在受体筛选技术的发展进程中,从Co-IP到CRISPR-Cas9都为布尼亚病毒的受体筛选贡献了力量。通过综合布尼亚病毒受体筛选中应用到手段及鉴定技术,循迹受体筛选技术的发展历程以及适用方向,结合候选受体在病毒生命进程中的作用机制,以帮助我们得以从布尼亚病毒的受体筛选中窥见技术发展的革新,以此来了解受体筛选未来可能的发展方向。本文将展示白纤病毒科、沙粒病毒科、汉坦病毒科、周布尼亚病毒科和内罗病毒科这五类布尼亚病毒中已经发现的主要受体类型,分析论述这些受体筛选时所用到的研究方法,以期为布尼亚病毒的受体研究提供一定的思路支持。
Bunyavirales of public health importance is an order of segmented negative⁃strand RNA viruses.containing more than 500 virus types Members of Bunyavirales can cause serious diseases in humans and other mammals due to their strong infectivity.The invasion mechanism of most bunyaviruses is not known,so targeted drugs and vaccines are lacking.A receptor study is a key step when analyzing the mechanism of virus infection and development of new drugs.Co⁃immunoprecipitation and clustered regularly interspaced short palindromic repeats(CRISPR)⁃CRISPR⁃associated protein 9(Cas9)have contributed to receptor screening for bunyaviruses.Integration of screening methods with identification of receptors is a rational approach for the development of new drugs.We describe the important receptors for the families Phenuiviridae,Arenaviridae,Hantaviridae,Peribunyaviridae and Nairoviridae to provide ideas for the receptor research of bunyaviruses.
