氮、钾对泰国葛愈伤组织次生代谢产物及生理生化的影响

摘要 目的:研究不同水平的氮、钾对泰国葛愈伤组织生理生化指标及次生代谢产物积累的影响。方法:利用植物生理学实验技术及HPLC法,对不同处理条件下的泰国葛愈伤组织生理生化指标及次生代谢产物积累规律进行探究。结果:泰国葛愈伤组织中葛根素、大豆苷、染料木素含量均与NH_(4)NO_(3)和KNO_(3)浓度呈负相关(P<0.01);NH_(4)NO_(3)和KNO_(3)浓度分别为1650.0、0.0 mg/L时,最有利于葛根素的生成;NH_(4)NO_(3)和KNO_(3)浓度分别为825.0、950.0 mg/L时,最有利于染料木素的生成;NH_(4)NO_(3)和KNO_(3)浓度分别为0.0、1900.0 mg/L时,最有利于大豆苷和总异黄酮的生成;葛根素含量与过氧化氢酶(CAT)活性、超氧化物歧化酶(SOD)活性呈负相关(P<0.05);大豆苷含量与丙二醛(MDA)含量呈正相关(P<0.01);染料木素含量与MDA含量、过氧化物酶(POD)活性呈正相关(P<0.05)。结论:不同水平的氮、钾对泰国葛愈伤组织中葛根素、大豆苷、染料木素的生成、MDA含量、SOD活性、POD活性、CAT活性均有一定的影响。 Objective:To study the physiological and biochemical index and accumulation of secondary metabolites in the callus of Pueraria mirifica affected by the different levels of nitrogen(N)and kalium(K).Methods:Physiological,biochemical and accumulation of secondary metabolites in the callus of P.mirifica under different treatment conditions were studied by means of plant physiology experiment and HPLC.Results:The contents of puerarin,daidzin and genistein in the callus of P.mirifica were negatively correlated with the concentrations of NH_(4)NO_(3) and KNO_(3)(P<0.01).The synthesis of puerarin was most favored when the concentrations of NH_(4)NO_(3) and KNO_(3) were 1650.0 mg/L and 0.0 mg/L,respectively.The synthesis of genistein was most favored when the concentrations of NH_(4)NO_(3) and KNO_(3) were 825.0 mg/L and 950.0 mg/L,respectively.When the concentrations of NH_(4)NO_(3) and KNO_(3) were 0.0 mg/L and 1900.0 mg/L,respectively,it was most favorable for the synthesis of daidzin and total isoflavones.The content of geranylgeranyl was negatively correlated with the activities of catalase(CAT)and superoxide dismutase(SOD)(P<0.05).The content of daidzin was positively correlated with the content of malondialdehyde(MDA)(P<0.01).The content of genistein was positively correlated with MDA content and peroxidase(POD)activity(P<0.05).Conclusion:The synthesis of puerarin,daidzin and genistein,MDA content,SOD activity,POD activity and CAT activity in the callus of P.mirifica are affected by different levels of N and K.
