摘要
共享镰孢菌Fusarium commune是引起莲腐败病的主要病原菌之一,明确其致病机理对于探寻莲腐败病防治策略具有重要意义。组蛋白乙酰转移酶在真核生物基因表达和各种生物过程的调控中起重要作用,然而,组蛋白乙酰转移酶FcHat1在共享镰孢菌的功能还未见报道。本研究利用BLASTP方法在共享镰孢菌全基因组数据中查找组蛋白乙酰转移酶Hat1同源蛋白,并分析FcHat1与同源蛋白的进化关系、保守基序和结构域等特征。根据同源重组的原理获得FcHat1-hph基因的融合片段,通过PEG介导的原生质体转化的方法获得FcHat1基因的敲除突变体和回补菌株,并对敲除突变体和回补菌株的主要生物学特性和致病力进行了分析。结果表明,FcHat1蛋白与其他同源蛋白具有相同的Hat1_N保守结构域,且与其他物种含Hat1_N结构域蛋白高度同源。对获得的FcHat1基因敲除突变体和回补菌株进行观察,结果表明敲除FcHat1不影响孢子的产量、孢子形态、孢子萌发以及DNA损伤胁迫的应答,但是ΔFcHat1突变体的生长速率、致病力与野生型相比显著降低,表明FcHat1在病菌营养生长、致病过程中发挥重要作用。
Fusarium commune is one of the main pathogens causing lotus rhizome rot,and it is important to clarify its pathogenesis to explore the control strategy of the disease.Histone acetyltransferases play important roles in the regulation of eukaryote gene expression and various biological processes;however,the function of histone acetyltransferase FcHat1 has not been reported in F.commune.In this study,BLASTP method was used to find histone acetyltransferase Hat1 homologous proteins in the whole genome data of F.commune,and the evolutionary relationship between FcHat1 and homologous proteins,and motifs,conserved domains and other features of FcHat1 were analyzed.Based on the principle of homologous recombination,a fusion fragment of FcHat1-hph gene was obtained.Knockout mutant and complementation strain of FcHat1 gene were obtained by PEG-mediated protoplast transformation,and the main biological characteristics and pathogenicity of the knockout mutant and complementation strain were analyzed.The results show that FcHat1 protein has the same Hat1_N conserved domain as other homologous proteins and is highly conserved to Hat1_N domain-containing proteins of other species.Knockout of FcHat1 did not affect conidial production,conidial morphology,conidial germination,and response to DNA damage stress,but the growth rate and pathogenicity of the ΔFcHat1 mutant were significantly reduced as compared with that of the wild type,indicating that FcHat1 played an important role in the vegetative growth and pathogenicity of the pathogen.
