摘要
选择邻苯二甲酸二(2-乙基己基)酯(DEHP)作为目标污染物,采用实验室前期从九溪人工湿地植物香蒲(Typha Linn)根际土中富集驯化分离的DEHP降解菌X41作为供试菌株,通过对其形态、生理生化特征及16S rDNA序列进行分析,初步鉴定菌株X41为蜡样芽孢杆菌(Bacillus cereus)。采用正交试验研究菌株X41对DEHP最佳降解条件和降解特性。结果表明,菌株X41在pH为6.0,温度为35℃,接菌量为3%,DEHP初始质量浓度为300 mg/L的环境条件下降解效率最高,且在此最优降解条件下,菌株X41生长良好,生长曲线呈“S”型,7 d的DEHP降解率高达99.54%。实验结果可为人工湿地中DEHP的去除提供一定的科学依据。
This study selected DEHP as the target pollutant and used DEHP degrading bacterium X41,which was enriched and domesticated from the rhizosphere soil of Typha Linn,a plant in the Jiuxi artificial wetland,as the test strain.By analyzing its morphology,physiological and biochemical characteristics,and 16S rDNA sequence,the strain X41 was preliminarily identified as Bacillus cereus.Orthogonal experiments were used to study the optimal degradation conditions and characteristics of strain X41 on DEHP.The results showed that strain X41 had the highest degradation efficiency under environmental conditions of pH 6.0,temperature 35℃,inoculation rate 3%,and initial DEHP concentration of 300 mg·L^(-1).And under this optimal degradation condition,strain X41 grows well,with a growth curve in an“S”shape,and a 7-day DEHP degradation rate of 99.54%.The results of this experiment can provide a certain scientific basis for the removal of DEHP in artificial wetlands.
