摘要
以中国被毛孢为发酵菌种,采用Box-Benhnken组合设计、响应面分析法对中国被毛孢胞外多糖(Ophiocordyceps sinensis extracellular polysaccharides,OPS)发酵的影响因素进行了优化研究,得出了最适液体发酵工艺:pH6.17、葡萄糖33.36 g/L、蚕蛹粉21.43 g/L,该条件下OPS含量可达355.79 mg/L,与理论值基本吻合。在此基础上,通过细胞增殖检测试剂盒测定OPS分别在100、200、400、600μg/mL浓度下对HT-29细胞的抑制率,发现其对HT-29细胞有较好的抑制作用。流式细胞仪检测表明,OPS对HT-29细胞生长的抑制作用与OPS诱导的细胞凋亡有关,细胞早期凋亡率从2.34%增加到24.0%。OPS处理后S期细胞增加到37.64%,G0/G1期细胞相应减少,OPS导致细胞周期在S期停滞。该研究结果可为OPS的开发利用提供新思路,为其在食品和药品中的开发与应用提供理论依据。
The optimal liquid fermentation process for Ophiocordyceps sinensis extracellular polysaccharides (OPS) fermentation was studied using Box Benhnken combination design and response surface analysis,using Ophiocordyceps sinensis as the fermentation strain.The optimal liquid fermentation process was found to be pH 6.17,glucose 33.36 g/L,and silkworm chrysalis powder 21.43 g/L.Under these conditions,the OPS content could reach 355.79 mg/L,which was consistent with the theoretical value.On this basis,the inhibition rates of OPS on HT-29 cells were measured using a cell proliferation detection kit at concentrations of 100,200,400,600 μg/mL,respectively.It was found that OPS had a good inhibitory effect on HT-29 cells.Flow cytometry analysis showed that the inhibitory effect of OPS on the growth of HT-29 cells was related to OPS induced apoptosis,with an increase in early cell apoptosis rate from 2.34% to 24.0%.After OPS treatment,the number of S-phase cells increased to 37.64%,and the number of G0/G1 phase cells decreased accordingly.OPS caused cell cycle arrest in the S-phase.This research result could provide new ideas for the development and utilization of OPS,and provide theoretical basis for its development and application in food and medicine.
