摘要
采用高效液相色谱法测定四妙勇安汤物质基准的指纹图谱及含量。使用ZORBAX SB-C_(18)色谱柱(250mm×4.6 mm,5μm),以乙腈-0.2%磷酸为流动相,梯度洗脱,流量为0.8 mL/min,检测波长为210 nm,柱温为25℃。建立15批四妙勇安汤物质基准的高效液相色谱指纹图谱,运用中药色谱指纹图谱相似度评价系统(国家药典委员会2012 A版)进行相似度评价,对共有峰进行药味归属,建立了绿原酸、哈巴苷、哈巴俄苷的含量测定方法。15批四妙勇安汤物质基准相似度均大于0.90,指纹图谱中含有11个共有峰,其中归属于金银花5个,玄参2个,当归2个,甘草2个。绿原酸、哈巴苷、哈巴俄苷平均质量分数分别为2.24%、0.59%、0.14%。建立的15批四妙勇安汤高效液相色谱指纹图谱精密度、稳定性良好,能够较好的反映其化学成分组成,可用于四妙勇安汤物质基准的质量控制与评价。
Fingerprint chromatograms and content of Simiao Yong'an Decoction substance benchmarks were determined by high performance liquid chromatography.ZORBAX SB-C_(18) column was used with acetonitrile-0.2%phosphoric acid aqueous solution as mobile phase with gradient elution,flow rate of 0.8 mL/min,detection wave length at 210 nm and column temperature at 25℃.15 batches of high performance liquid chromatography fingerprint chromatogram of Simiao Yong'an decoction were set up.The Similarity Evaluation System for Chromatographic Fingerprint of TCM(State Pharmacopoeia Commission 2012 A version)was applied for similarity evaluation,and the common peaks were identified and assigned.The method was established for the determination of the contents of chlorogenic acid,harpagide,harpagoside.The similarity of the 15 batches of high performance liquid chromatography fingerprint chromatogram of Simiao Yong'an decoction established was greater than 0.90,the 11 common peaks were identified and assigned.in which 5 peaks belonged to Honeysuckle,2 peaks belonged to Scrophularia ningpoensis,2 peaks belonged to Angelica Sinensis,2 peaks belonged to Glycyrrhiza uralensis.The average content of chlorogenic acid,harpagide,harpagoside was 2.24%,0.59%,0.14%,respectively.The high performance liquid chromatography fingerprint chromatograms set up in this study was precise,stable.It can reflect the chemical components of this formula,and the method can be used for its quality control and evaluation.
