摘要
窄叶性状作为水稻理想株型育种的重要指标之一,直接影响光合作用,进而影响水稻的生物学产量。福建漳浦野生稻具有明显的窄叶特性,但其窄叶性状的分子调控机制尚未明晰。研究以福建漳浦野生稻与宽叶型水稻R527连续回交得到的稳定遗传的窄叶型株系S为材料,以亲本R为对照,进行转录组分析和表型性状测定。结果发现,S株系的剑叶宽度明显变窄,千粒重显著下降,剑叶长度和稻谷粒大小未发生明显变化。通过转录组测序获得650个差异表达基因,其中381个基因上调表达,269个基因下调表达,对其中9个差异基因进行RT-qPCR的分析,结果表明9个差异基因的表达趋势与转录组测序结果基本一致。差异基因主要参与植物激素、DNA损伤修复等方面的代谢通路。WD40蛋白基因均在窄叶株系中上调表达,而大部分糖基转移酶却下调表达,由此推测WD40蛋白和糖基转移酶可能协同影响植物激素、DNA损伤修复等代谢途径调控漳浦野生稻窄叶的形成。
As a critical factor in developing ideal-type rice variety,narrow leaves directly affect photosynthesis leading to the biological yield of rice.The Zhangpu wild rice has an obvious narrow-leaf trait,but the molecular regulation mechanism is still unclear.Using the Zhangpu wild rice as the raw material,a backcross introduced line population with R527 background was constructed.Narrow leaf lines were analyzed using transcriptome technology.The results showed that the blade width and 1000-grain weight of the narrowleaf lines were decreased significantly,while the blade length and grain size were not changed significantly.The results of transcriptome sequencing showed that 650 differentially expressed genes were found in the narrow-leaf lines,of which 381 were up-regulated and 269 down-regulated.In which,the relative c of the 9 differentially expressed genes using the RT-qPCR was basically consistent with the transcriptome sequencing results.The differentially expressed genes were mainly involved in the metabolic pathways such as plant hormones and DNA damage repair.The differentially expressed genes related WD40 proteins were upregulated in narrow-leaf lines,while most of differentially expressed genes related glycosyl transferase were down-regulated.It was speculated that the WD40 proteins and glycosyl transferases might synergistically regulate the formation of narrow leaves in Zhangpu wild rice by affecting plant hormones,DNA damage repair or other metabolic pathways.
