基于UPLC-Q-TOF-MS/MS结合网络药理学及分子对接探究桑叶清肺润燥的潜在药效成分和作用机制

摘要 目的本研究以中医药理论中桑椹滋阴补血的功效为依据,利用超高压液相色谱串联四级杆飞行时间质谱(UPLC-TOF-MS/MS)技术结合网络药理学探讨桑叶清肺润燥的潜在药效成分和作用机制。方法利用UPLC-TOF-MS/MS技术结合SwissADME鉴定桑叶的主要活性成分,使用PharmMapper数据库、Swiss Target Prediction数据库预测桑叶活性成分的靶点,在GeneCards数据库、SymMap数据库中筛选肺热咳嗽和肺燥咳嗽的潜在作用靶点。将成分靶点与症状群靶点取交集后,导入STRING数据库构建PPI网络获得核心靶点,最后对核心靶点基因做GO生物过程分析和KEEG通路富集分析。结果结果显示桑椹有效药效成分有55个,靶点261个,与肺热咳嗽、肺燥咳嗽证候相关的核心靶点分别为15和27个。四个证候群的GO生物过程和KEGG通路分析结果存在共性和特性,HIF1x信号通路、白细胞跨内皮迁移是桑叶清肺润燥的主要途径。结论因此桑叶通过多成分、多靶点、多通路的协同作用发挥清肺润燥作用,为进一步研究探讨桑椹滋阴补血的药效成分及作用机制研究提供了理论依据。 Objective This study is based on the effect of nourishing Yin and replenishing blood of mulberry in traditional Chinese medicine theory,to investigate the potential pharmacodynamic components and mechanism of Folium mori in clearing lung-heat and moisturizing drynness by using ultra high pressure liquid chromatography-time-off-flight mass spectrometer(UPLC-TOF-MS/MS)combined with network pharmacology.Methods The main active components of Folium mori were identified by UPLC-TOF-MS/MS technique combined with SwissADME.PharmMapper database and Swiss Target Prediction database was used to predict the targets of active components.And the potential targets in treating lung heat cough and lung dryness cough were screened in GeneCards database and SymMap database.The component target was intersected with the symptom cluster target,and then the intersection was imported into STRING database to construct PPI network to obtain the core targets.Finally,GO biological process analysis and KEEG pathway enrichment analysis were performed on the core target genes.Results The results showed that there were 55 effective components and 261 targets of Folium mori,and there were 15 core targets related to lung heat cough and 27 core targets related to lung dryness cough syndrome.The results of GO biological process and KEGG pathway analysis of the four syndrome groups had commonality and characteristics.HIF1x signaling pathway and leukocyte transendothelial migration were the main pathways of Folium mori clearing lung-heat and drying dryness.Conclusions Folium mori plays a role in clearing lung-heat and moistening dryness through the synergistic action of multi-components,multi-targets and multi-pathways,which provides a theoretical basis for further research on the pharmacodynamic components and mechanism of Folium mori in nourishing Yin and tonifying blood.
