摘要
目的探讨金合欢素调节胰岛素受体底物-1(IRS-1)/磷脂酰肌醇3激酶(PI3K)/丝氨酸苏氨酸蛋白激酶(AKT)信号通路对糖尿病大鼠胰岛素抵抗(IR)的影响。方法选取6周龄,体质量220~235 g的SPF级SD雄性大鼠60只作为研究对象。随机选出12只大鼠作为对照组(NC组),其余48只大鼠构建2型糖尿病(T2DM)大鼠模型,造模成功后随机分为糖尿病组、金合欢素组、MG53组、金合欢素+MG53组,每组12只。检测所有大鼠空腹血糖(FBG)、总胆固醇(TC)、低密度脂蛋白(LDL)、甘油三脂(TG)水平。采用酶联免疫吸附试验检测空腹胰岛素(FINS)水平以及胰腺组织白细胞介素(IL)-1β、肿瘤坏死因子-α(TNF-α)水平,计算胰岛素敏感指数(ISI)和胰岛素抵抗指数(HOMA-IR);计算口服葡萄糖耐量实验曲线下面积(AUC);采用苏木精和伊红(HE)染色检测胰腺组织病理变化;采用Western blot法检测IRS-1/PI3K/AKT通路蛋白表达水平。结果与NC组相比,糖尿病组大鼠体质量、FBG、TC、TG、LDL水平均明显升高,差异均有统计学意义(P<0.05);与糖尿病组相比,金合欢素组大鼠体质量、FBG、TC、TG、LDL水平均明显下降,而MG53组均明显升高,差异均有统计学意义(P<0.05);与金合欢素组相比,金合欢素+MG53组大鼠体质量、FBG、TC、TG、LDL水平均明显升高,差异均有统计学意义(P<0.05)。与NC组相比,糖尿病组FBG、FINS、HOMA-IR水平均明显升高,AUC明显增大,ISI水平明显下降,差异均有统计学意义(P<0.05);与糖尿病组相比,金合欢素组FINS、HOMA-IR水平均明显下降,AUC明显减小,ISI水平明显升高,而MG53组FINS、HOMA-IR水平均明显升高,AUC明显增大,ISI水平明显下降,差异均有统计学意义(P<0.05);与金合欢素组相比,金合欢素+MG53组FINS、HOMA-IR水平均明显升高,AUC明显增大,ISI水平明显下降,差异均有统计学意义(P<0.05)。与NC组相比,糖尿病组TNF-α、IL-1β水平均明显升高,差异均有统计学意义(P<0.05);与糖
Objective To investigate the effect of acacetin on insulin resistance(IR)in diabetic rats by regulating insulin receptor subset-1(IRS-1)/phosphatidylinositol 3 kinase(PI3K)/serine-threonine protein kinase(AKT)signaling pathway.Methods A total of 60 SPF SD male rats,aged 6 weeks and weighing 220-235 g,were selected as the study subjects.Twelve rats were randomly selected as the control group(NC group),and the rest 48 rats were used to establish type 2 diabetes mellitus(T2DM)rat models.After successful modeling,the rats were randomly divided into diabetes group,acacetin group,MG53 group,and acacetin+MG53 group,with 12 rats in each group.The levels of fasting blood glucose(FBG),total cholesterol(TC),low density lipoprotein(LDL)and triglyceride(TG)were detected.The levels of fasting insulin(FINS),interleukin(IL)-1βand tumor necrosis factor-α(TNF-α)in pancreatic tissue were detected by enzyme-linked immunosorbent assay.Insulin sensitivity index(ISI)and insulin resistance index(HOMA-IR)were calculated.The area under the curve(AUC)of oral glucose tolerance test was calculated.Hematoxylin-eosin(HE)staining was used to detect the pathological changes of pancreatic tissue.The protein expression of IRS-1/PI3K/AKT pathway was detected by Western blot.Results Compared with the NC group,the body weight,FBG,TC,TG and LDL levels of the diabetic group were significantly increased(P<0.05).Compared with the diabetic group,the body weight,FBG,TC,TG and LDL levels of the acacetin group were significantly decreased(P<0.05),while those of the MG53 group were significantly increased(P<0.05).Compared with the acacetin group,the acacetin+MG53 group had significant increases in the body weight,FBG,TC,TG,and LDL levels(P<0.05).Compared with NC group,the levels of FBG,FINS and HOMA-IR in DM group were significantly increased,AUC was significantly increased,and ISI was significantly decreased,with statistically significant differences(P<0.05).Compared with the diabetic group,the levels of FINS,HOMA-IR and AUC in the acacetin group decrease
