基于16S rRNA测序研究金振口服液对支气管炎豚鼠肠道菌群的影响

Study on the Effect of Jinzhen Oral Liquid on the Intestinal Flora of Guinea Pigs with Bronchitis Based on 16S rRNA Sequencing

摘要 目的:研究金振口服液对支气管炎豚鼠肠道菌群的调节作用。方法:实验动物随机分为空白组、模型组、金振口服液(JZOL)高、中、低剂量组和阿莫西林组,共6组。除空白组外,其余各组豚鼠通过脂多糖(LPS)和辣椒素诱导制备气管炎模型。给药后观察豚鼠一般状态和咳嗽典型症状,处死豚鼠后剪开盲肠收集肠内容物。采用HE染色法观察肺组织病理变化,16S rRNA基因测序检测肠道菌群变化。结果:JZOL高、中、低剂量组均能改善豚鼠的一般状态,减少咳嗽次数;肺组织病理结果显示,JZOL高、中、低剂量组肺组织病理改变较模型组均有所减轻,其中JZOL高剂量组病理变化最低;16S rRNA测序结果显示,Chao1,ACE和Shannon指数在各组之间无明显差异,各给药组对肠道菌群多样性无显著影响,阿莫西林组与各组肠道菌群结构显著分离;在门水平上,不同剂量的JZOL均降低了造模后升高的拟杆菌门丰度,其中JZOL高剂量组还增加了造模后降低的疣微菌门和变形菌门丰度。在属、种水平上,造模后嗜黏蛋白阿克曼菌丰度降低,而JZOL恢复了嗜黏蛋白阿克曼菌丰度。LEfSe分析显示,JZOL中、低剂量组较模型组相比,瘤胃球菌、类阿氏动丝菌、帕拉普氏菌等丰度也明显增加。结论:金振口服液可能通过调节肠道菌群结构,进而改善支气管炎症。 Objective:To study the regulation effect of Jinzhen oral liquid on intestinal flora of guinea pigs with bronchitis(JZOL).Methods:The experimental animals were randomly divided into 6 groups,blank group,model group,JZOL high,medium,and low dose groups,and amoxicillin group.Except the blank group,all the other groups were given lipopolysaccharide(LPS)and capsaicin to prepare bronchitis model.After administration,the general state and typical cough symptoms of guinea pigs were observed.After the guinea pigs were euthanized,the cecum was cut and the intestinal contents were collected.HE staining was used to observe the pathological changes of lung tissue,and 16S rRNA gene sequencing was used to detect the changes of intestinal flora.Results:JZOL high,medium,and low dose groups can all improve the general state of guinea pigs and reduce the frequency of coughing.The pathological results of lung tissue showed that the pathological changes of lung tissue in the JZOL high,medium,and low dose groups were alleviated compared to the model group,with the lowest pathological changes observed in the JZOL high group.16S rRNA sequencing results showed that Chao1,ACE and Shannon indexes were not significantly different among all groups,and each administration group had no significant effect on intestinal flora diversity,while the intestinal flora structure of amoxicillin group was significantly separated from that of each group.At the phylum level,JZOL of different doses all reduced the abundance of increased Bacteroidetes after modeling,at the same time,high-dose JZOL also reversed the abundance of Verrucomicrobia and Proteobacteria which decreased after modeling.At the genus and species levels,the abundance of Akkermansiaceae decreased after modeling,while JZOL restored the abundance of Akkermansiaceae.LEfSe analysis showed that,compared to the model group,Ruminococcus,Kineothrix alysoides and Paraprevotella also showed significant enrichment in the JZOL medium and low dose groups.Conclusions:Jinzhen oral liquid may improve bro
