基于加权基因共表达网络鉴定食管癌浸润性淋巴细胞关键节点基因

Identification of Key Hub Genes in Infiltrating Lymphocytes of Esophageal Carcinoma Based on Weighted Gene Co-expression Network

摘要 目的分析食管癌肿瘤浸润性淋巴细胞抗原受体区的基因表达,利用加权基因共表达网络分析(weighted gene co-expression network analysis,WGCNA)方法筛选食管癌的关键基因,为相关研究提供数据支撑。方法选择安阳市肿瘤医院70例食管癌患者的组织标本进行基因测序,同时,从NCBI SRA数据库下载部分转录组数据。通过R语言的WGCNA模块构建食管癌共表达网络,并鉴别与食管癌关联性最强的共表达模块,关键基因由Centiscape筛选。结果网络分析的结果表明,MEgrey60、MEpurple和MEblack模块与食管癌高度相关。依据蛋白—蛋白相互作用关系,本研究从每个模块中筛选了关联度最高的20个关键基因。这些模块中的基因主要涉及内肽酶抑制剂活性和特异性RNA聚合酶Ⅱ转录因子活性、组蛋白结合、抗氧化活性、硫氧还蛋白过氧化物酶活性、ATP酶活性和蛋白质转运蛋白活性等生物学功能。结论本研究筛选出的食管癌浸润性淋巴细胞的关键基因,可为食管癌的诊断、治疗和免疫治疗提供靶点和理论依据。 Objective To use Weighted Gene co-expression Network Analysis(WGCNA)identifing key genes associated with esophageal cancer and provide data support for related research,in order to identify the gene expression of the infiltrating lymphocyte antigen receptor region in esophageal cancer.Methods Tissue samples of 70 esophageal cancer patients from our hospital was selected and gene sequenced,meanwhile,transcriptome data were downloaded from NCBI SRA database.The co-expression network of esophageal cancer was constructed by R WGCNA,and the co-expression modules that most strongly correlated with esophageal squamous cell carcinoma were screened out.The hub genes were analyzed by Centiscape.Results The results of network analysis indicate that MEgrey60、MEpurple and MEblack modules were highly correlated with esophageal cancer.According to protein-protein interaction,the top 20 hub genes of each module were selected.The biological function of the genes in the module are mainly involved in endopeptidase inhibitor activity,specific RNA polymeraseⅡtranscription factor activity,histone binding,antioxidant activity,thioredoxin peroxidase activity,ATPase activity and protein transporter activity.Conclusion The key genes of infiltrating lymphocytes in esophageal cancer identified in this study can provide targets and theoretical basis for the diagnosis,treatment,and immunotherapy of esophageal cancer.
