摘要
目的:观察三焦针法对原发性失眠大鼠下丘脑组织谷氨酸(glutamic acid,Glu)、γ-氨基丁酸能神经元(gamma-aminobutyric acid,GABA)表达的影响,探讨三焦针法治疗PI的作用机制。方法:SD雄性大鼠随机分为空白组、模型组、地西泮组和三焦组,每组6只。除正常组外,其余各组采用“PCPA腹腔注射方式”建立PI大鼠模型。地西泮组予地西泮灌胃(0.9 mg/kg),三焦组针刺膻中、中脘、气海、血海(双侧)及足三里(双侧),诸穴行捻转补法30 s,1次/d,均治疗14 d。观察大鼠一般状态;测定大鼠体质量;Elisa法检测下丘脑组织Glu、GABA表达。结果:与空白组相比,PI模型大鼠一般状态较差,体质量显著降低(P<0.01),下丘脑组织中Glu表达水平显著升高(P<0.01)、GABA表达水平显著降低(P<0.01)。与模型组比较,地西泮组、三焦组大鼠体质量显著升高(P<0.01,P<0.05),下丘脑组织中Glu表达水平显著降低(P<0.01)、GABA表达水平显著升高(P<0.01),地西泮组与三焦组比较无统计学意义(P>0.05)。结论:三焦针法可改善PI大鼠失眠症状,这可能与调节下丘脑组织中Glu、GABA表达水平有关。
Objective To observe the effects of San Jiao acupuncture on the expression of Glu(Glutamic Acid,Glu)、GABA(Gamma-aminobutyric Acid,GABA)in the hypothalamus of rats with primary insomnia(PI),and to explore the mechanism of San Jiao acupuncture in the treatment of PI.Methods SD male rats were randomly divided into blank group,model group,diazepam group and Sanjiao group,6 rats in each group.Except for the normal group,the other groups used“PCPA intraperitoneal injection method”to establish PI rat model.In the diazepam group,diazepam was administered by gavage(0.9 mg/kg),and in the Sanjiao group,danzhong,zhongkou,qihai,blood sea(bilateral)and zhousanli(bilateral)were needled,and the acupuncture points were twisted and tonicized for 30 s,once/d,and the treatment was carried out for 14 d.Observations were made on the general state of the rats,the body mass of the rats was determined,and the expression of Glu and GABA in hypothalamus tissues was detected by the Elisa assay.Results Compared with the blank group,the general state of PI model rats was worse,body mass was significantly lower(P<0.01),and the expression levels of Glu(P<0.01)and GABA(P<0.01)in the hypothalamus tissues were significantly higher(P<0.01)and lower(P<0.01).Compared with the model group,the body mass of rats in diazepam group and Sanjiao group was significantly higher(P<0.01,P<0.05),and the expression level of Glu in hypothalamic tissue was significantly lower(P<0.01)and higher(P<0.01)than that of GABA,and there was no statistically significant difference in the comparison of diazepam group and Sanjiao group(P>0.05).Conclusion Sanjiao acupuncture can improve insomnia symptoms in PI rats,which may be related to the regulation of the expression levels of Glu and GABA in the hypothalamic tissues.
