基于网络药理学探讨郁金散治疗猪肠道炎症的作用机制

Study on the Mechanism of Yujin Powder in the Treatment of Porcine Intestinal Inflammation Based on Network Pharmacology

摘要 为了利用网络药理学探讨郁金散治疗猪肠道炎症的作用靶点,并对靶点涉及的生物学过程和作用通路进行阐述,本试验借助中药系统药理学数据库(TCMSP)分别筛选郁金散的活性成分和靶点,利用美国国家生物技术信息中心(NCBI)、在线人类孟德尔遗传(OMIM)和GeneCards数据库检索肠道炎症靶点,通过Uniprot数据库对靶点进行校正。利用Cytoscape 3.9.1软件和String网站分别构建共有靶点网络和核心靶点相互作用网络,利用用于注释、可视化和集成发现的数据库(DAVID)对靶点涉及的生物学过程和信号通路进行分析。结果显示:经选择物种“Sus scrofa”校正后,郁金散的活性成分有66个,靶点81个,猪肠道炎症靶点198个,郁金散治疗猪肠道炎症的预测靶点有38个,其中核心靶点25个,包括肿瘤坏死因子(TNF)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)等。基因本体论(GO)功能富集分析得到生物学过程(BP)条目91个,细胞组成(CC)条目3个,分子功能(MF)条目10个,其中生物学过程主要涉及免疫反应、RNA聚合酶II启动子的转录正调控和细胞对脂多糖的反应等;细胞组成主要涉及胞外区、细胞外间隙和白细胞介素-6受体复合体;分子功能主要涉及细胞因子活性、生长因子活性和白细胞介素-2受体结合等。京都基因与基因组百科全书(KEGG)通路富集分析得到83条通路,相关性较强的通路依次是癌症通路、南美锥虫病和细胞因子-细胞因子受体相互作用等。结果表明,郁金散通过调控TNF、IL-6和IL-10等靶点,参与癌症通路、南美锥虫病和细胞因子-细胞因子受体相互作用等通路,对猪肠道炎症发挥治疗作用。 To explore the targets of Yujin powder in treating porcine intestinal inflammation using network pharmacology,and to elucidate the biological processes and pathways involved,this study utilized the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)to screen the active components and targets of Yujin powder.Intestinal inflammation targets were retrieved from the National Center for Biotechnology Information(NCBI),Online Mendelian Inheritance in Man(OMIM),and GeneCards databases,and corrected using the UniProt database.The common target network and core target interaction network were constructed using Cytoscape 3.9.1 software and the String website.The Database for Annotation,Visualization,and Integrated Discovery(DAVID)was used to analyze the biological processes and signaling pathways involving the targets.The results showed that,after correcting for the species"Sus scrofa",there were 66 active components and 81 targets of Yujin powder,198 targets for porcine intestinal inflammation,and 38 predicted targets for Yujin powder in treating porcine intestinal inflammation,with 25 core targets including tumor necrosis factor(TNF),interleukin-6(IL-6),and interleukin-10(IL-10).Gene Ontology(GO)functional enrichment analysis identified 91 biological process(BP)entries,3 cellular component(CC)entries,and 10 molecular function(MF)entries.The biological processes mainly involved immune response,positive regulation of transcription from RNA polymerase II promoter,and cellular response to lipopolysaccharide.The cellular components mainly involved the extracellular region,extracellular space,and interleukin-6 receptor complex.The molecular functions mainly involved cytokine activity,growth factor activity,and interleukin-2 receptor binding.Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis identified 83 pathways,with the most relevant pathways being cancer pathways,Chagas disease,and cytokine-cytokine receptor interaction.
