摘要
目的探讨间隙连接蛋白B3(GJB3)对肺腺癌细胞H1299增殖、迁移和侵袭能力的影响。方法利用RT-qPCR和Western blot实验分别检测人正常肺上皮细胞(BEAS-2B)和肺腺癌细胞系(H441、A549、H1299)中GJB3的mRNA和蛋白水平的表达;筛选出GJB3表达量最高的H1299细胞系作为后续实验研究对象;对H1299细胞进行小干扰RNA(siRNA)转染,分为对照组(si-NC)和实验组(si-GJB3),在孵育0、24、48、72、96 h后采用CCK-8法检测H1299细胞活力,使用平板克隆检测细胞增殖能力,细胞划痕实验检测细胞的迁移能力,Transwell实验检测细胞的侵袭能力。结果与人正常肺上皮细胞相比,在肺腺癌细胞中GJB3的mRNA和蛋白的表达量均较高(P<0.05),其中H1299细胞的表达量最为显著(P<0.05);在H1299细胞中敲低GJB3后,明显抑制了该细胞的活力、增殖能力和迁移及侵袭能力(P<0.05)。结论GJB3在肺腺癌细胞中高表达,可能促进肺腺癌细胞的发生和发展。
Objective To investigate the effects of gap connexin B3(GJB3)on the proliferation,migration and invasion of lung adenocarcinoma cell H1299.Methods The mRNA and protein levels of GJB3 in human nor-mal lung epithelial cells(BEAS-2B)and lung adenocarcinoma cell lines(H441,A549,H1299)were detected by RT-qPCR and Western blot assay,respectively.The H1299 cell line with the highest expression of GJB3 was selected as the object of follow-up experiment.H1299 cells were transfected with small interfering RNA(siRNA)and divided into control group(si-NC)and experimental group(si-GJB3).The H1299 cells viability were detected by CCK-8 after incubation for 0 h,24 h,48 h,72 h and 96 h.The proliferation ability of the cell was detected by plate cloning,the migration ability of the cells were detected by cell scratch assay,and the invasion ability of the cell was detected by Transwell assay,respectively.Results Compared with human normal lung epithelial cells,the expression levels of GJB3 mRNA and protein was higher in lung adenocarcinoma cells,and the expression levels of H1299 cells were the most significant(P<0.05).After knocking down GJB3 in H1299 cells,the activity migration and invasion ability proliferation ability of the cells was significantly inhibited compared with the control group(P<0.05).After knockdown of GJB3 in H1299 cells,the of the cells was significantly inhibited.Knockdown of GJB3 in H1299 cells significantly inhibited the of the cells(P<0.05).Conclusion The expression of GJB3 in lung adenocarcinoma cells is higher than that in normal lung epithelial cells.Knockdown of GJB3 may promote the occurrence and development of lung adenocarcinoma cells.
