过敏原对过敏性鼻炎患者外周血CD4^(+)Th1 IL-18、IL-18BPa和IL-18Rα表达的影响

摘要 目的探讨过敏原对健康人(healthy control,HC)和过敏性鼻炎(allergic rhinitis,AR)患者外周血CD4^(+)Th1细胞表达IL-18、IL-18BPa和IL-18Rα蛋白水平及mRNA水平的影响。方法收集皮肤点刺试验阴性的鼻炎(AR^(-))、皮肤点刺试验阳性的鼻炎(AR^(+))患者和HC的外周血标本。流式细胞术检测过敏原对CD4^(+)Th1细胞表达IL-18、IL-18BPa和IL-18Rα蛋白水平的影响;qPCR检测过敏原对CD4^(+)T细胞表达IL-18、IL-18BPa和IL-18RαmRNA水平的影响。结果与HC相比,AR^(-)和AR^(+)患者外周血Th1细胞中IL-18蛋白的表达增加,IL-18BPa蛋白的表达减少;AR^(+)患者外周血Th1细胞中IL-18Rα蛋白的表达增加;过敏原可增强HC Th1细胞IL-18Rα蛋白的表达,并诱导AR^(+)患者和HC CD4^(+)T细胞IL-18、IL-18BPa和IL-18RαmRNA的表达。结论过敏原极可能通过促进外周血CD4^(+)Th1细胞表达IL-18和IL-18Rα进而参与AR发病。 Objective To investigate the effects of allergens on the expressions of IL-18,IL-18BPa and IL-18Rαprotein in peripheral blood CD4^(+)Th1 cells of healthy control subjects(HC)and patients with allergic rhinitis(AR),and on the expressions of IL-18,IL-18BPa and IL-18Rα mRNA in the peripheral blood CD4^(+)T cells.Methods Blood samples were collected from patients with rhinitis for negative skin prick test(AR^(-)),rhinitis for positive skin prick test(AR^(+))and HC.Flow cytometry was used to evaluate the effects of allergens on the expressions of IL-18,IL-18BPa and IL-18Rαprotein in CD4^(+)Th1 cells.The expressions of IL-18,IL-18BPa and IL-18RαmRNA in CD4^(+)T cells were determined by qPCR.Results Compared with HC,increased IL-18 while decreased IL-18BPa expressions in Th1 cells of AR^(-)and AR^(+)patients were observed,increased IL-18Rαexpression in Th1 cells of AR^(+)patients was also found.Additionally,allergens induced elevated expression of IL-18Rα protein in Th1 cells of HC,and induced elevated mRNA expressions of IL-18,IL-18BPa and IL-18Rα in isolated blood CD4^(+)T cells of AR^(+)patients and HC.Conclusion Allergens may be involved in the pathogenesis of AR by inducing the expressions of IL-18 and IL-18Rαin blood CD4^(+)Th1 cells.
