摘要
以癸酸插入率为指标,采用混料设计对酶法合成中长链甘三酯(MLCT)的3种Sn-1,3位特异性脂肪酶(Lipozyme RM IM、Lipozyme TL IM和NS40086)配比进行优化,并对MLCT产物的脂肪酸组成及复合酶的稳定性进行分析。结果表明:3种酶对MLCT产物癸酸插入率的影响依次为NS40086>Lipozyme RM IM>Lipozyme TL IM,Lipozyme RM IM、Lipozyme TL IM和NS40086的最佳质量比为0.323∶0.122∶0.555,在此条件下MLCT产物的癸酸插入率为46.87%±0.24%,Sn-2位油酸含量为73.13%,Sn-1,3位癸酸含量为62.60%,制得的复合酶具有良好的稳定性。
Using decylic acid insertion rate as index,the ratio of three Sn-1,3 specific lipase(Lipozyme RM IM,Lipozyme TL IM and NS40086)was optimized by mixture design for enzymatic synthesis of medium and long chain triglycerides(MLCT),and the fatty acid composition of MLCT products and stability of compound lipase were analyzed.The results showed that the effects of three lipase on decylic acid insertion rate of MLCT products were NS40086>Lipozyme RM IM>Lipozyme TL IM.The optimal mass ratio of Lipozyme RM IM,Lipozyme TL IM and NS40086 was 0.323:0.122:0.555.Under the conditions,the decylic acid insertion rate was 46.87%±0.24%,the Sn-2 oleic acid content was 73.13%,and the Sn-1,3 decylic acid content of MLCT products was 62.60%.The obtained compound lipase had good stability.
