不同NDF水平开食料对羔羊小肠发育及IGF-1、IGF-1R基因表达的影响

Effects of Starter Diets with Different NDF Levels on The Development of Small Intestine and The Expression of IGF-1 and IGF-1R Genes in Lambs

摘要 为研究不同中性洗涤纤维(NDF)水平的开食料对湖羊羔羊小肠发育以及胰岛素样生长因子-1(IGF-1)和胰岛素样生长因子-1受体(IGF-1R)基因表达的影响。试验选取健康的湖羊公羔54只,随机分为三组,每组18只羊,单栏饲养,分别饲喂15%、20%、25%NDF水平的开食料,试验从7日龄开始至56日龄时屠宰取样,测定,用实时荧光定量PCR方法检测空肠、回肠中上述基因的相对表达水平。结果表明,饲喂不同NDF水平的开食料对羔羊空肠和回肠的重量与长度无显著影响(P>0.05),对羔羊空肠和回肠的相对重量与相对长度无显著影响(P>0.05)。在空肠中IGF-1基因的相对表达量25%NDF水平组显著高于15%NDF水平组(P<0.05),NDF水平为25%与20%两组差异极显著(P<0.01),NDF水平为15%与20%两组差异不显著(P>0.05),IGF-1R基因的相对表达量20%NDF水平组与15%和25%NDF水平组差异极显著(P<0.01),NDF水平为15%与25%两组差异不显著(P>0.05)。在回肠中IGF-1和IGF-1R基因的相对表达量15%NDF水平组极显著高于20%和25%NDF水平组(P<0.01),NDF水平为20%与25%两组差异不显著(P>0.05)。结果显示,开食料对羔羊空肠、回肠的长度和重量无显著影响,在空肠中25%NDF水平组显著上调了IGF-1基因的表达,20%NDF水平组上调了IGF-1R基因的表达;在回肠中15%NDF水平可以显著上调IGF-1和IGF-1R基因的表达。 To investigate the impact of varying levels of neutral detergent fiber(NDF)in dietary intake on the maturation of lamb small intestine and the expression patterns of insulin-like growth factor-1(IGF-1)and insulin-like growth factor-1 receptor(IGF-1R)genes,an in-depth examination was conducted.54 healthy Hu sheep lambs were selected for the experiment and randomly allocated into three groups,with 18 sheep in each group.They were individually housed and provided with starters containing NDF levels of 15%,20%,and 25%respectively.At the age of 56 days,they were euthanized for sampling,and the relative expression levels of the aforementioned genes in both jejunum and ileum were determined using a real-time fluorescence quantitative PCR method.The results indicate that the provision of starters containing varying levels of NDF did not yield a statistically significant impact on the weight and length of the jejunum and ileum in lambs(P>0.05),nor did it have a significant effect on the relative weight and relative length of these segments in lambs(P>0.05).In the jejunum,the relative expression level of the IGF-1 gene was significantly higher in the group with a 25%NDF level compared to the group with a 15%NDF level(P<0.05),and there was a highly significant difference between the groups with a 25%NDF level and those with a 20%NDF level(P<0.01).However,no significant difference was observed between the groups with a 15%NDF level and those with a 20%NDF level(P>0.05).The relative expression level of the IGF-1R gene exhibited a highly significant difference between the group with a 20%NDF level and both the groups with a 15%and 25%NDF levels(P<0.01),but no significant difference was found be‐tween the groups with a 15%and those with a 25%NDF levels(P>0.05).In the jejunum,the relative ex‐pression levels of IGF-1 and IGF-1R genes were significantly higher in the group with a 15%NDF level compared to those in the groups with 20%and 25%NDF levels(P<0.01),while no significant difference was observed between the groups with 2
