L-苏糖酸镁对组胺和氯喹诱导的小鼠瘙痒敏化的抑制作用

摘要 目的:探究L-苏糖酸镁(L-TAMS)对组胺和氯喹引起的小鼠急性瘙痒及瘙痒敏化的影响。方法:将56只小鼠随机平均分为7组,对照组(A组)、组胺瘙痒敏化组(B组)、氯喹瘙痒敏化组(C组)、L-TAMS+组胺瘙痒敏化组(D组)、L-TAMS+氯喹瘙痒敏化组(E组)、组胺瘙痒敏化+L-TAMS组(F组)、氯喹瘙痒敏化+L-TAMS组(G组)。A组生理盐水注射对照,B、D、F组采用组胺(2 mg/ml,50μl)连续注射4天瘙痒敏化造模,C、E、G组采用氯喹(2 mg/ml,50μl)连续注射4天瘙痒敏化造模;D、E组在敏化造模前预防性口服L-TAMS一周,F、G组小鼠在敏化后口服L-TAMS一周。每次注射组胺和氯喹后进行瘙痒行为学测试。结果:与B组相比,D组第1天注射组胺后的搔抓次数没有统计学意义(P>0.05);D组第3、4天注射组胺后的搔抓次数明显减少(P<0.01)。与B组相比,D、F组第11天注射组胺后的搔抓次数明显减少(P<0.001)。与C组相比,E组第1天注射氯喹后的搔抓次数没有统计学意义(P>0.05);E组第4天注射氯喹后的搔抓次数明显减少(P<0.01)。与C组相比,E、G组第11天注射氯喹后的搔抓次数明显减少(P<0.05,P<0.01)。结论:口服L-TAMS能有效预防和逆转小鼠被组胺和氯喹诱导的瘙痒敏化。 Objective:To explore the effects of L-Threonate Magnesium(L-TAMS)on acute itching and sensitization induced by histamine and chloroquine in mice.Methods:56 mice were randomly divided into 7 groups:Control(Group A),Histamine Sensitization(Group B),Chloroquine Sensitization(Group C),L-TAMS+Histamine Sensitization(Group D),L-TAMS+Chloroquine Sensitization(Group E),Histamine Sensitization+L-TAMS(Group F)and Chloroquine Sensitization+L-TAMS(Group G).Group A received saline injections as a control.Groups B,D and F were sensitized with histamine(2 mg/ml,50μl)injections for 4 days,while Groups C,E and G with chloroquine(2 mg/ml,50μl)for 4 days.Groups D and E were pre-treated with L-TAMS for one week before sensitization and Groups F and G after sensitization.Pruritus behavior tests were conducted after each histamine and chloroquine injection.Results:Compared with Group B,Group D showed no significant difference in scratching frequency on the first day(P>0.05),but a significant reduction on the third and fourth days(P<0.01)and a marked reduction on the eleventh day(P<0.001).Compared with Group C,Group E showed no significant difference on the first day(P>0.05),but a significant reduction on the fourth day(P<0.01)and a marked reduction on the eleventh day(P<0.05,P<0.01)for both Groups E and G.Conclusion:Oral L-TAMS effectively prevents and reverses itch sensitization induced by histamine and chloroquine in mice.
