摘要
目的研究1株黄河三角洲滨海湿地来源的真菌Fusarium oxysporum RD-79的次级代谢产物及其生物活性。方法利用硅胶柱色谱、凝胶柱色谱、半制备高效液相色谱等分离方法对该菌株发酵产物进行分离纯化,通过核磁共振(NMR)、质谱(MS)等方法,结合相关文献对比鉴定化合物结构;分别采用CCK8法和Griess法对化合物进行抗肿瘤活性和抗炎活性测试。结果从Fusarium oxysporum RD-79的发酵产物中分离得到7个单体化合物,分别为5-butyl-4-methoxy-6-methyl-2H-pyran-2-one(1)、delitpyrone D(2)、3-hydroxymethyl-6,8-dimethoxycoumaarin(3)、5-[(1S,2S)-1,2-dihydroxybutyl]-4-methoxy-6-methyl-2H-pyran-2-one(4)、setosphapyrone A(5)、5-(1-hydroxybutyl)-4-methoxy-6-methyl-2H-pyran-2-one(6)、9-O-methylscytalol A(7)。结论化合物1~7均可不同程度抑制NO的产生;在质量浓度为100μg/mL下,化合物1~7均无抗肿瘤活性。
Objective To study the secondary metabolites and their biological activities of a fungus Fusarium oxysporum RD-79 from the coastal wetland of the Yellow River Delta.Methods The fermentation products of the strain were separated and purified by silica gel column chromatography,ODS column chromatography,semi-preparative high performance liquid chromatography and other separation methods.Their structures were elucidated based on the spectral data of nuclear magnetic resonance analyses(NMR)and mass spectrometry(MS)and compared with literature data.CCK8 and Griess methods were used to test the cytotoxic activity and anti-inflammatory activity of the compounds.Results Seven pure compounds were isolated and identified as 5-butyl-4-methoxy-6-methyl-2H-pyran-2-one(1),delitpyrone D(2),3-hydroxymethyl-6,8-dimethoxycoumaarin(3),5-[(1S,2S)-1,2-dihydroxybutyl]-4-methoxy-6-methyl-2H-pyran-2-one(4),setosphapyrone A(5),5-(1-hydroxybutyl)-4-methoxy-6-methyl-2H-pyran-2-one(6),9-O-methylscytalol A(7),respectively.Conclusion Compounds 1-7 could inhibit the production of NO in varying degrees.At the concentration of 100μg/mL,compounds 1-7 showed no anti-tumor activity.
