白鲜皮-苦参致Wistar大鼠肝损伤的微生物-脂代谢机制

摘要 目的:通过高通量测序技术检测白鲜皮-苦参致Wistar大鼠肝损伤后的微生物变化,探讨对白鲜皮-苦参致肝损伤的潜在微生物机制。方法:雌性Wistar大鼠随机分为正常组、中药低、中、高剂量组4组,以白鲜皮-苦参不同剂量(生药量分别为4.125、8.25、16.5 g·kg^(-1))灌胃28 d,记录一般情况,计算肝脏体质量比、观测血清生化指标、苏木素-伊红(HE)染色观察肝脏病理学改变、16S rDNA高通量测序检测大鼠粪便微生物变化。结果:与正常组比较,白鲜皮-苦参给药后Wistar大鼠肝脏重量和肝脏体质量比增加,中药中、高剂量组肝脏重量差异有统计学意义(P<0.05),中药低、中、高剂量组肝脏体质量比均具有统计学意义(P<0.05)。与正常组比较,中药中、高剂量组血清白蛋白、胆固醇水平增加(P<0.05)。中药中、高剂量组肝脏组织病理学出现微小空泡样改变。与正常组比较,白鲜皮-苦参给药后出现明显的肠道菌群紊乱,中药中、高剂量组α多样性增加。主坐标分析显示,随着给药剂量的增加,物种越来越偏离正常组。与正常组比较,给药后厚壁菌门/拟杆菌门比例下降,属水平的Parasutterella、 Romboutsia、 Turicibacter、 Allobaculum、 Dubosiella升高,Lachnospiraceae_NK4A136_group、 Blautia、Erysipelatoclostridium、Muribaculum、Ruminococcus_gnavus_group降低。相关性分析表明,Parasutterella、Romboutsia、Turicibacter、Allobaculum、Dubosiella与血清胆固醇、肝脏体质量比正相关,lanchnospiraceae_NK4A136_group、Blautia、Muribaculum、Erysipelatoclostridium、Ruminococcus_gnavus_group与血清胆固醇、肝脏体质量比负相关。结论:白鲜皮-苦参的肝损伤表现为脂代谢紊乱,其机制与脂代谢相关微生物增加有关。 Objective: To observe the microbial changes in Wistar rats with liver injury caused by the Dictamni Cortex-Flavescens Sophora by high-throughput sequencing technology and investigate the potential mechanism of liver injury caused by the Dictamni Cortex-Flavescens Sophora. Method: Female Wistar rats were randomly divided into four groups: normal group, as well as low-dose, medium-dose, and high-dose groups of traditional Chinese medicine(TCM). The rats were gavaged with the Dictamni Cortex-Flavescens Sophora in different doses(4.125, 8.25, 16.5 g·kg^(-1) of raw drug respectively) for 28 days, and the general condition was recorded. The liver-body weight ratio was calculated, and the biochemical indexes of serum were observed. The Hematoxylin-eosin(HE) staining was used to observe pathological changes in the liver, and 16S rDNA high-throughput sequencing was utilized to detect fecal microbial changes in rats. Result: Compared with the normal group, Dictamni Cortex-Flavescens Sophora increased the liver weights and liver-body weight ratios of Wistar rats. The difference in liver weight between the medium-dose and high-dose groups of TCM was statistically significant(P<0.05), and the liver-body weight ratios of the low-dose, medium-dose, and highdose groups of TCM were all statistically significant(P<0.05). Compared with the normal group, serum albumin and cholesterol levels increased in the medium-dose and high-dose groups of TCM(P<0.05). The histopathology of the liver in the medium-dose and high-dose groups of TCM showed tiny vacuole-like changes.Compared with the normal group, there were obvious intestinal flora disorders after administration of Dictamni Cortex-Flavescens Sophora, and alpha diversity increased in the medium-dose and high-dose groups of TCM.The principal coordinates analysis showed that species increasingly deviated from the normal group as the administered dose increased.
