摘要
【目的】比较双峰驼(Camelus bactrianus)正常睾丸及隐睾中性激素受体分布差异,探究双峰驼隐睾病理学变化与性激素受体的相关性,为双峰驼睾丸病理研究提供形态学参考。【方法】采用常规组织切片染色及特殊染色法观察隐睾病理学组织形态以及糖原和蛋白的分布变化;应用免疫组织化学法及免疫荧光技术定位雄激素受体(androgen receptor,AR)、雌激素受体α(estrogen receptor alpha,ERα)、雌激素受体β(estrogen receptor beta,ERβ)、促黄体生成素受体(luteinizing hormone receptor,LHR)和促卵泡激素受体(follicle-stimulating hormone receptor,FSHR)在正常睾丸及隐睾的分布及表达。【结果】与正常睾丸相比,隐睾中各类细胞数量显著减少,Leydig细胞发育迟缓,Sertoli细胞多呈幼稚态,结缔组织及生精小管基膜酸性黏蛋白阳性强度下降,总蛋白分布下降;ERα、LHR和FSHR的表达极显著升高(P<0.01),AR的表达极显著降低(P<0.01),ERβ的表达差异不显著(P>0.05);AR在Leydig细胞和Sertoli细胞中的表达强度减弱,ERα在Sertoli细胞中的表达强度减弱,ERβ在Leydig细胞和Sertoli细胞中的表达强度无变化,LHR和FSHR在Leydig细胞和Sertoli细胞中的表达强度增强。【结论】双峰驼隐睾组织结构发育迟缓,糖原及蛋白分泌异常。隐睾Leydig细胞和Sertoli细胞性激素受体表达异常,不利于精子正常发生,阻碍生精细胞发育。
[Purpose]To compare the distribution of sex hormone receptors between normal testis and cryptorchidism of Camelus bactrianus,and to explore the correlation between the pathological changes of cryptorchidism and sex hormone receptors,providing morphological reference for the study of testicular pathology in C.bactrianus.[Methods]The histopathological morphology of cryptorchidism and the distribution of glycogen and protein were observed by routine tissue section staining and special staining.The distribution and expression of androgen receptor(AR),estrogen receptor alpha(ERα),estrogen receptor beta(ERβ),luteinizing hormone receptor(LHR)and follicle-rsity(Natural Science)E-mail:ynauzkxb@foxmail.com stimulating hormone receptor(FSHR)in normal testis and cryptorchidism were detected by immunohistochemistry and immunofluorescence.[Results]Compared with normal testis,the number of various types of cells in the cryptorchidism was significantly lower,the development of Leydig cells was slow,and Sertoli cells were mostly immature,the positive intensity of acid mucin in connective tissue and the basement membrane of seminiferous tubules decreased,and the total protein distribution decreased;the expression of ERα,LHR and FSHR in cryptorchidism was extremely significantly higher(P<0.01),the expression of AR was extremely significantly lower(P<0.01),and there was no significant difference in the expression of ERβ(P>0.05);the expression intensity of AR in Leydig cells and Sertoli cells decreased,the expression intensity of ERαin Sertoli cells decreased,the expression intensity of ERβin Leydig cells and Sertoli cells did not change,and the expression intensity of LHR and FSHR in Leydig cells and Sertoli cells increased.[Conclusion]The cryptorchidism tissue structure of C.bactrianus develop slowly,and the glycogen and protein secretion is abnormal.The abnormal expression of sex hormone receptors in Leydig cells and Sertoli cells of cryptorchidism is not conducive to the normal occurrence of sperm and hinders the developmen
