摘要
用于白及多糖(Bletilla striata polysaccharide,BSP)提取主要有溶剂提取法、辅助提取法等;分离纯化主要有活性炭或双氧水脱色,Sevage法脱蛋白,柱色谱法分离等;含量测定中通常使用苯酚-硫酸法和蒽酮-硫酸法;分子量采用高效液相色谱法、红外光谱、凝胶渗透色谱等方法来进行测定;结构表征主要采用液相色谱,红外光谱或核磁共振法来进行测定。白及多糖为葡甘聚糖,具有抗氧化、抗炎、抗肿瘤、免疫调节和促进伤口愈合等多种生物活性,在生物医药领域有广泛的应用前景。BSP有良好的药理活性,目前其研究大部分为粗多糖,药理活性仍在细胞阶段。因此对其提取分离纯化、结构鉴定、生物活性的研究仍是社会关注的热点。主要综述了BSP的提取分离纯化、结构组成及生物活性的研究进展并对其进行系统分析,为BSP的深度开发利用以及后续的研究提供了理论基础。
The main methods used for extracting Baiji(Bletilla striata)polysaccharide(BSP)include solvent extraction and auxiliary extraction.The main methods of separation and purification include activated carbon or hydrogen peroxide decoloriza⁃tion,Sevage deproteinization and column chromatography separation.Phenol sulfuric acid method and anthrone sulfuric acid method are commonly used in content determination.The molecular weight was determined by high performance liquid chroma⁃tography,infrared spectroscopy,gel permeation chromatography and other methods.Structural characterization is mainly deter⁃mined using liquid chromatography,infrared spectroscopy or nuclear magnetic resonance methods.BSP is a polysaccharide that has various biological activities such as antioxidant,anti-inflammatory,anti-tumor,immune regulation and promoting wound healing effects.It has broad application prospects in the field of biomedicine.BSP has good pharmacological activity,and cur⁃rently most of its research is on crude polysaccharides,and its pharmacological activity is still in the cellular stage.Therefore,the research on its extraction,separation,purification,structural identification and biological activity is still a hot topic of social con⁃cern.This article mainly reviewed the research progress in the extraction,separation,purification,structural composition and bio⁃logical activity of BSP and systematically analyzed it,providing a theoretical basis for the deep development and utilization of BSP and subsequent research.
