摘要
将体质量(127.07±10.92)g的中华绒螯蟹(Eriocheir sinensis,以下简称河蟹)成体雄蟹和体质量(86.16±6.80)g雌蟹离水,分别暴露于室温(20~22℃)下0 h(对照组)、8 h、16 h和24 h,随机选取12只蟹(雌、雄各6只)采肝胰腺和肌肉样品,测定组织匀浆清液中琥珀酸脱氢酶(SDH)、乳酸脱氢酶(LDH)、苹果酸脱氢酶(MDH)和过氧化氢酶(CAT)的活性及丙酮酸、丙二醛(MDA)含量、总抗氧化能力(T-AOC)和肝/肌糖原比,探讨不同干露胁迫时间对河蟹成体呼吸代谢、能量代谢及抗氧化能力的影响。结果显示:(1)雄蟹肝胰腺中LDH活性随干露时间的延长呈先下降后上升趋势,在24 h时最高;雌、雄蟹肌肉中LDH活性均随干露时间延长而下降,而SDH活性呈先上升后下降趋势,在8 h时最高;(2)雄蟹肝胰腺中的糖原含量随干露时间的延长而显著上升;雌、雄蟹肝胰腺中的丙酮酸含量先下降后上升,在24 h最高;雄蟹肌肉中的糖原含量在24 h时最高,而肌肉中MDH活性显著下降;(3)雌蟹和雄蟹肝胰腺中的T-AOC和CAT活性均在24 h时最高,显著高于其他时间点;雌蟹肝胰腺MDA含量在8 h时最高,而雄蟹肝胰腺中的MDA含量在24 h时最高;肌肉中的MDA含量、T-AOC和CAT活性随干露时间的延长而下降。综上所述,雌、雄蟹的抗干露胁迫能力存在差异,24 h内的干露胁迫显著影响河蟹成体的呼吸代谢和抗氧化能力,但能量代谢基本不变,因此河蟹可通过调节自身的生理代谢来适应24 h的离水运输。
To investigate the effects of different air exposure stress periods on respiratory metabolism,energy metabolism and antioxidant capacity of adult Chinese mitten handed crab(Eriocheir sinensis),The male crab with body weight of(127.07±10.92)g and female crab with body weight of(86.16±6.80)g were exposed to air for 0 h(control group),8 h,16 h or 24 h,and 12 crabs at room temperature(20-22℃),and then the hepatopancreas and muscle were sampled in 6 males and 6 females for determination of the indices of respiratory metabolism,energy metabolism and antioxidant capacity,including activities of lactate dehydrogenase(LDH),succinate dehydrogenase(SDH),malate dehydrogenase(MDH)and catalase(CAT),contents of pyruvate,and malondialdehyde(MDA),total antioxidant capacity(T-AOC)and liver/muscle glycogen ratio,The results showed that(1)the activity of hepatopancreas lactate dehydrogenase(LDH)in male crabs was decreased first and then increased with increase in air exposure period,with the maximal value at 24 h.There was significant decrease in activity of LDH in muscle of both male and female crab,while the activity of succinate dehydrogenase(SDH)was found to be first increased and then decreased,with the maximal level in 8 h.(2)The glycogen content in the hepatopancreas of male crab was shown to be significantly increased with increase in air exposure period.The pyruvate content in hepatopancreas of both male and female crabs was decreased first and then increased,with the maximal level at 24 h,and the maximal glycogen content in the muscle of male crab in 24 h,while the activity of muscle malate dehydrogenase showed a significantly decreasing trend.(3)The maximal T-AOC and CAT activities were observed in the hepatopancreas of female and male crabs at 24 h,significantly higher than those at other time,the maximal malondialdehyde(MDA)content in the hepatopancreas of female crabs in 8 h,and the maximal hepatopancreas MDA content of male crabs in 24 h.The muscle MDA content,T-AOC and CAT activities were decreased with increase
