钙离子胁迫对菘蓝叶片生理特性的影响

摘要 目的为解析钙离子胁迫对菘蓝叶片生长的影响,以菘蓝为实验材料,观察了钙离子胁迫对叶片的影响。方法以蛭石为基质,在上口直径为7 cm的小花盆中培养菘蓝种子,浇施霍格兰德营养液,培养条件为温度25℃、光照强度3600μmol/(m^(2)·s)、相对湿度为75%。当长出6片叶时,用含不同钙离子浓度(0、200、400 mmol/L)的霍格兰德营养液处理菘蓝植株,观察光合特性及其他生理指标变化规律。结果与对照相比,Ca^(2+)处理下菘蓝叶片净光合速率(Pn)、蒸腾速率(Tr)、气孔导度(Gs)、胞间CO_(2)浓度(Ci)、叶绿素a、叶绿素b、总叶绿素显著降低,含水量逐渐下降;总可溶性糖含量与脯氨酸含量随着钙离子浓度的增加逐渐上升,甜菜碱的含量先升高后降低;水分利用率(WUE)、电导率、丙二醛(MDA)含量、过氧化氢含量、超氧阴离子含量随着钙离子浓度的增加呈升高趋势;超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX)、单脱氢抗坏血酸还原酶(MDHAR)、脱氢抗坏血酸还原酶(DHAR)和谷胱甘肽还原酶(GR)活性随着钙离子浓度的增加呈升高趋势。结论表明钙离子胁迫能明显抑制菘蓝叶片的光合特性,并对叶片产生渗透胁迫。菘蓝叶片可能通过合成渗透保护物质、调节渗透压,通过抗氧化酶系统清除活性氧。 Objective In order to analyze the effects of calcium ion stress on the growth of leaves of Isatis indigotica,the effect of calcium ion stress on leaves of I.indigotica was observed.Methods The seeds of the I.indigotica were grown based on vermiculite in small pots with 7cm diameter at the upper mouth and watered with Hoagland nutrient solution under the culture conditions of 25℃,3600μmol/(m^(2)·s)light intensity and 75%relative humidity.When the plant grows out six leaves,the plants were treated with Hoagland nutrient solution containing calcium ions at different concentrations(0,200,400 mmol/L)and the changes of photosynthetic characteristics and other physiological indexes were observed.Results The results showed that,compared with control group,the net photosynthetic rate(Pn),transpiration rate(Tr),stomatal conductance(Gs),intercellular CO_(2) concentration(Ci),and the content of chlorophyll a,chlorophyll b,total chlorophyll and water was decreased gradually under calcium treatment.The content of total soluble sugar and proline was increased gradually with the increase of calcium ion concentration,while the content of betaine was increased first and then decreased.Water utilization efficiency(WUE),electrolyte leakage rate,malondialdehyde(MDA)content,hydrogen peroxide content and superoxide anion content was increased with the increase of calcium ion concentration.Activity of the superoxide dismutase(SOD),peroxidase(POD),catalase(CAT),ascorbate peroxidase(APX),monodehydroascorbate reductase(MDHAR),dehydroascorbate reductase(DHAR)and glutathione reductase(GR)was increased with the increase of calcium ion concentration.Conclusion These suggest that calcium stress can inhibit the photosynthetic properties of leaves of I.indigotica significantly.It also causes osmotic stress on I.indigotica leaves.I.indigotica leaves may regulate osmotic pressure by synthesizing osmotic protective substances,and remove reactive oxygen species through antioxidant enzyme system.
