摘要
目的:探究表皮生长因子受体突变体Ⅲ(EGFRvⅢ)与丙酮酸激酶亚型2(PKM2)在乳腺癌中表达的相关性。方法:选择2020年8月—2023年7月期间我院手术治疗的乳腺癌患者60例作为研究对象,通过免疫组化等实验检测EGFRwt、EGFRvⅢ、磷酸化EGFR、PKM2、β-连环蛋白(β-catenin)和磷酸化的β-catenin在乳腺癌和癌旁组织之间的表达差异,利用Spearman法相关性分析各变量间的相关性,从而验证乳腺癌中EGFRwt/vⅢ与PKM2的关系。结果:癌组织EGFRwt、EGFRvⅢ、磷酸化EGFR、PKM2、β-catenin磷酸化的β-catenin平均光密度值均高于癌旁组织(P<0.05)。癌组织EGFRwt、EGFRvⅢ、磷酸化EGFR、PKM2、β-catenin磷酸化的β-catenin表达阳性率均高于癌旁组织(P<0.05)。EGFRwt与EGFRvⅢ、磷酸化EGFR、PKM2、β-catenin、磷酸化的β-catenin均呈正相关(r=0.865、0.754、0.691、0.625、0.678,P<0.05),EGFRvⅢ与磷酸化EGFR、PKM2、β-catenin、磷酸化的β-catenin均呈正相关(r=0.582、0.564、0.817、0.756,P<0.05),PKM2与β-catenin、磷酸化的β-catenin均呈正相关(r=0.524、0.631,P<0.05)。结论:EGFRwt/vⅢ与PKM2的表达呈正相关。这一发现可能有助于进一步理解乳腺癌的发生机制,并为乳腺癌的诊断和治疗提供新的靶点和策略。
Objective:To explore the correlation between epidermal growth factor receptor variantⅢ(EGFRvⅢ)and pyruvate kinase M2(PKM2)expression in breast cancer.Methods:A total of 60 breast cancer patients who underwent surgical treatment at our hospital from August 2020 to July 2023 were selected as the study subjects.Immunohistochemistry and other experiments were performed to detect the expression differences of EGFRwt,EGFRvⅢ,phosphorylated EGFR,PKM2,β-catenin,and phosphorylatedβ-catenin between breast cancer tissue and adjacent non-cancerous tissue.The Spearman correlation analysis was used to evaluate the correlation between variables,aiming to validate the relationship between EGFRwt/vⅢand PKM2 in breast cancer.Results:The average optical density values of EGFRwt,EGFRvⅢ,phosphorylated EGFR,PKM2,β-catenin,and phosphorylatedβ-catenin in cancer tissue were significantly higher than those in adjacent non-cancerous tissue(P<0.05).The positive expression rates of EGFRwt,EGFRvⅢ,phosphorylated EGFR,PKM2,β-catenin,and phosphorylatedβ-catenin in cancer tissue were also higher than those in adjacent non-cancerous tissue(P<0.05).EGFRwt showed positive correlations with EGFRvⅢ,phosphorylated EGFR,PKM2,β-catenin,and phosphorylatedβ-catenin(r=0.865,0.754,0.691,0.625,0.678;P<0.05).EGFRvⅢexhibited positive correlations with phosphorylated EGFR,PKM2,β-catenin,and phosphorylatedβ-catenin(r=0.582,0.564,0.817,0.756;P<0.05).PKM2 showed positive correlations withβ-catenin and phosphorylatedβ-catenin(r=0.524、0.631;P<0.05).Conclusion:There is a positive correlation between EGFRwt/vⅢand PKM2 expression in breast cancer.This finding may contribute to a better understanding of the mechanisms underlying breast cancer and provide new targets and strategies for its diagnosis and treatment.
