摘要
目的 研究流苏石斛的黄酮类成分及其对皮质酮诱导的PC12细胞损伤的保护作用。方法采用聚酰胺树脂、葡聚糖凝胶、硅胶柱色谱等方法进行成分分离,通过核磁共振波谱分析与鉴定结构;采用体外细胞实验对所得化合物进行活性检测。结果 从流苏石斛中分离得到10个黄酮及其苷类成分,分别鉴定为新西兰牡荆苷Ⅱ(1)、异佛莱心苷(2)、佛莱心苷(3)、芹菜素-6-C-α-L-鼠李糖-8-C-β-D-异鼠李糖苷(4)、芹菜素-6-C-α-L-阿拉伯糖-8-C-β-L-阿拉伯糖苷(5)、芹菜素-6,8-二-C-α-L-阿拉伯糖苷(6)、金圣草黄素-7-O-β-D-葡萄糖苷(7)、芹菜素-6-C-α-L-鼠李糖苷(8)、异牡荆素(9)和芹菜素(10),其中化合物3~6、8~10可显著提高皮质酮诱导损伤的PC12细胞的存活率。结论 化合物4、5、7~10为首次从该种植物中分离得到,化合物5、7、8为首次从该属植物中分离得到;化合物3~6、8~10对皮质酮诱导的PC12细胞损伤具有一定的神经保护作用。
Objective To determine flavonoid components from Dendrobium fimbriatum and its protective effect on PC12 cell damage induced by corticosterone.Methods The compounds were separated and purified by polyamide resin,Sephadex LH-20 column chromatography,and silica gel column chromatography.The structures of the isolated compounds were analyzed and identified with nuclear magnetic resonance spectroscopy.The activity of the compounds was tested by in vitro cell assay.Results Totally 10 flavonoids and their glycosides were isolated from Dendrobium fimbriatum,and identified as vicenin-2(1),isoviolanthin(2),violanthin(3),apigenin-6-C-α-L-rhamnosyl-8-C-β-D-quinovoside(4),apigenin-6-C-α-L-arabinosyl-8-C-β-D-arabinoside(5),apigenin-6,8-di-C-α-L-arabinoside(6),chryseriol-7-O-β-D-glucoside(7),apigenin-6-C-α-L-rhamnoside(8),isovitexin(9),and apigenin(10).Compounds 3~6 and 8~10 significantly improved the survival rate of PC12 cell damage induced by corticosterone.Conclusion Compounds 4,5,and 7~10 have been isolated from Dendrobium fimbriatum for the first time.Compounds 5,7 and 8 have been isolated from Dendrobium for the first time.Compounds 3~6 and 8~10 show neuroprotective effect on PC12 cell damage induced by corticosterone.
