摘要
胚胎干细胞具有显著的增殖和分化发育能力,因此在细胞治疗、疾病建模和药物研发方面具有巨大的潜力。但高效的全能性却导致其往往伴随基因组的不稳定性,尤其在DNA损伤方面更加凸显。人类胚胎干细胞拥有特殊的修复机制,用以避免这类损伤在细胞内的积累或向子代的传递,进而保障自我复制和分化发育的准确性和安全性。本文对人类胚胎干细胞DNA损伤的修复机制进行了总结,旨在为结合其修复机制,进一步探索干细胞治疗的新思路提供依据。
Embryonic stem cells have remarkable proliferative and differentiation developmental capabilities and thus have great potential for cell therapy,disease modeling,and drug development.However,such totipotency has resulted in genomic instability,especially in the case of DNA damage.Human embryonic stem cells possess particular repair mechanisms to avoid the accumulation of such damage within the cell or its transmission to the offspring,thus ensuring the accuracy and safety of self-replication and differentiation development.This paper summarized the repair mechanisms of DNA damage in human embryonic stem cells and intends to provide a basis for further exploration of new ideas of stem cell therapy in conjunction with their repair mechanisms.
