摘要
旨在探究胆汁酸(BAs)对亚急性瘤胃酸中毒(subacute ruminal acidosis,SARA)山羊瘤胃发酵和菌群结构的影响。试验选取8只生长期雄性波杂山羊安装永久性瘤胃瘘管,康复后随机平均分为SARA组和SARA+BAs组,预饲期为2周,试验期8周,预饲期内由精粗比50∶50的日粮转为70∶30的高精料日粮诱导SARA,每只添加胆汁酸10 g/d,于试验期第1、14、28、42、56天晨饲后0、2、3、4和6 h进行瘤胃液pH值的动态监测,在第56天晨饲前采集瘤胃液,检测乳酸、挥发性脂肪酸及菌群结构。结果显示:SARA组山羊平均日增重显著升高(P<0.05),饲料转化率有降低的趋势(P<010);SARA组山羊瘤胃液pH<58的持续时间大于3 h,处于SARA状态;而SARA+BAs组瘤胃液pH<58的时间均少于3 h;与SARA组相比,SARA+BAs组瘤胃内乙酸浓度显著升高(P<0.05),而乳酸浓度显著降低(P<0.05)。菌群分类学分析显示,SARA+BAs组Chao1指数、Shannon指数和菌门数量均显著高于SARA组(P<0.05);在门水平上,SARA+BAs组脱硫菌门(Desulfobacterota)相对丰度显著升高(P<0.05),而疣微菌门(Verrucomicrobiota)、绿弯菌门(Chloroflexi)相对丰度显著降低(P<0.05);在属水平上,SARA+BAs组毛螺菌科_NK3A20群(Lachnospiraceae_NK3A20_group)、互营球菌属(Syntrophococcus)相对丰度显著降低(P<0.05),而瘤胃球菌科_NK4A214(Ruminococcaceae_NK4A214)、未分类的颤螺菌科(Norank_f_Oscillospiraceae)、韦荣球菌科UCG-001(Veillonellaceae_UCG-001)的相对丰度显著升高(P<0.05)。研究表明,日粮添加胆汁酸可提升山羊瘤胃菌群的丰富度和多样性,改善瘤胃发酵,从而缓解高精料日粮诱导的SARA。
The aim of this study was to evaluate the effect of feeding bile acids(BAs)on ruminal fermentation,and microbial community in goats under subacute ruminal acidosis(SARA)induced by high concentrate diets.Eight goats during growth stage fitted with permanent rumen fistula were randomly divided into two groups:the SARA group and the SARA+BAs group.A high-concentrate diet with a ratio of con-centrate to forage of 70∶30 was used to induce SARA,and the dose of BAs supplementation was 1 g/day.The pre-feeding period lasted for 2 weeks,followed by an 8 weeks experimental period.Ruminal pH in the goats was dynamically monitored at 0,2,3,4,and 6 hours after morning feeding on experimental days 1,14,28,42,and 56,respectively.The levels of lactate,volatile fatty acids,and microbial composi-tion in the ruminal fluid of the animals were measured on D56.The results showed that the average daily gain was significantly higher in the SARA+BAs group than in the SARA group(P<0.05),but the feed conversion ratio tended to decrease(P<010).Their ruminal pH below 58 lasted more than 3 hours after morning feeding on days 1,14,28,42,and 56 in the SARA group.However,the dietary supplementation with BAs obviously increased the ruminal pH in the goats,which showed a shorter duration(less than 3 h)of rumen pH<58.Compared with the SARA group,the runimal acetate concentration in the SARA+BAs group was significantly increased,while their lactate concentration was markedly decreased by BAs treatment(P<0.05).The microbial community analysis revealed that the Chao1 index,Shan-non index,and the number of bacterial phyla in the SARA+BAs group were significantly higher than those in the SARA group(P<0.05).At the phylum level,Desulfobacterota phylum abundance in the goats was significantly increased(P<0.05),while the Verrucomicrobiota and Chloroflexi phyla level were significantly decreased(P<0.05)by BAs.At the genus level,the relative abundances of Lachnospiraceae_NK3A20_group and Syntrophococcus were significantly decreased(P<0.05),while the relative
