摘要
目的:探讨黄连散对2,4-二硝基氯苯(DNCB)诱导的特应性皮炎(AD)模型小鼠的治疗作用。方法:将42只雄性BALB/c小鼠,随机分为正常组、模型组、氢化可的松组、黄连散油剂低、中、高剂量(0.3、0.6、1.2 g·kg^(-1))组、黄连散水提液(0.6 g·kg^(-1))组。除正常组外,其余组均于小鼠背部涂抹DNCB以建立AD模型。在DNCB刺激后的第15天各组给予相应药物或溶剂,观察并记录小鼠皮损变化、皮炎评分、抓挠次数;苏木素-伊红(HE)染色观察皮肤和脾脏组织病理学改变;实时荧光定量聚合酶链式反应(Real-time PCR)检测皮肤中丝聚蛋白(FLG)、兜甲蛋白(LOR)、内披蛋白(IVL)及脾脏免疫球蛋白E(lgE)、白细胞介素-4(IL-4)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)和γ干扰素(IFN-γ)的mRNA水平。结果:与正常组比较,模型组小鼠皮肤出现红肿、结痂等症状,皮炎评分、抓挠次数和脾脏指数均上升(P<0.05),皮肤组织中FLG、LOR、IVL的表达水平显著降低(P<0.01),脾脏中IgE、IL-4、IL-6、IL-1β、TNF-α的mRNA表达量显著升高,IFN-γ的表达量显著降低(P<0.01);与模型组比较,各药物组小鼠皮肤红斑、鳞屑及结痂等症状均有不同程度减轻,皮炎评分、抓挠次数及脾脏指数有所下降(P<0.05,P<0.01);此外,药物组小鼠皮肤中FLG、LOR、IVL的表达水平升高(P<0.05,P<0.01),脾脏中IgE、IL-4、IL-6、IL-1β、TNF-α的mRNA表达量减少,IFN-γ升高(P<0.05,P<0.01),皮肤和脾组织病变情况均有不同程度改善,以黄连散油剂中剂量组和氢化可的松组最为明显(P<0.05,P<0.01);且黄连散油剂中剂量组各指标均优于黄连散水提液组。结论:黄连散可能通过上调皮肤屏障蛋白表达水平,抑制辅助性T细胞2(Th2)相关炎症因子表达,提高辅助性T细胞1(Th1)炎症因子表达,恢复皮肤屏障功能及脾脏中Th1/Th2平衡,抑制AD小鼠脾脏组织炎症反应,从而缓解AD;黄连散油剂比水剂
Objective:To explore the therapeutic effect of Huangliansan on atopic dermatitis(AD)model mice induced by 2,4-dinitrochlorobenzene(DNCB).M ethod:A total of 42 male BALB/c mice were randomly divided into normal group,model group,hydrocortisone group,low,medium,and high-dose groups(0.3,0.6,1.2 g·kg^(-1))of Huangliansan oil,and water extract group(0.6 g·kg^(-1))of Huangliansan.In addition to the normal group,DNCB was applied on the back of mice in other groups to establish the AD model.On the 15thday after DNCB stimulation,each group was given the corresponding drug or solvent,and the changes in skin lesions,dermatitis score,and frequency of scratching were observed and recorded.Hematoxylin-eosin(HE)staining was used to observe the histopathological changes in the skin and spleen.Real-time fluorescent quantitative polymerase chain reaction(Real-time PCR)was used to detect mRNA levels of filaggrin(FLG),lorophane(LOR),and involucrin(IVL)in skin,as well as immunoglobulin E(lgE),interleukin-4(IL-4),interleukin-6(IL-6),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),and interferon-γ(IFN-γ)in spleen.Result:Compared with the normal group,the model group showed symptoms of skin swelling and scab,and the score of dermatitis,the frequency of scratching,and the spleen index were increased(P<0.05).The expression levels of FLG,LOR,and IVL in skin tissue were significantly decreased(P<0.01),and the mRNA expressions of IgE,IL-4,IL-6,IL-1β,and TNF-αin the spleen were significantly increased,while the expression level of IFN-γwas significantly decreased(P<0.01).Compared with the model group,the symptoms of skin erythema,scaly,and scab of mice in each drug group were alleviated to varying degrees,and the score of dermatitis,the frequency of scratching,and the spleen index were decreased(P<0.05,P<0.01).In addition,the expression levels of FLG,LOR,and IVL in the skin of mice in the drug group were increased(P<0.05,P<0.01),and the mRNA expression of IgE,IL-4,IL-6,IL-1β,and TNF-αin spleen were decreased.IFN-γwas increa
