基于网络药理学探讨麻杏化瘀汤治疗儿童肺炎支原体肺炎的作用机制

To explore the mechanism of action of Maxing Huayu Tang(麻杏化瘀汤)on mycoplasma pneumoniae pneumonia in children based on network pharmacology

摘要 目的基于网络药理学的方法探究麻杏化瘀汤主要成分治疗儿童肺炎支原体肺炎(MPP)的可能分子机制。方法在中药系统药理学分析平台(TCMSP)中检索并提取麻杏化瘀汤的有效活性成分及相关作用靶点;在疾病靶点筛选数据库Genecards和OMIM中筛选出儿童MPP的相关基因,与药物作用靶点取交集,筛选出麻杏化瘀汤治疗儿童MPP的潜在靶点,并构建药物-化学成分-关键靶点网络图;采用STRING数据库和Cytoscape软件绘制蛋白相互作用(PPI)网络;对关键靶点进行基因本体(gene ontology,GO)富集分析和京都基因与基因组百科全书(kyotoencyclopedia of genes and genomes,KEGG)通路富集分析。结果筛选出麻杏化瘀汤有效活性成分共126个,药物靶点与疾病靶点的交集靶点共78个。GO富集得到了632个条目,包括生物过程497个、分子功能91个、细胞组分44个。KEGG富集分析得到相关通路134个,主要与胞内磷脂酰肌醇激酶(PI3K)/蛋白激酶B(Akt)信号通路、丝裂原活化蛋白激酶(MAPK)信号通路、白细胞介素-17(IL-17)、肿瘤坏死因子(TNF)信号通路、T细胞17(Th17)细胞分化有关。结论本研究基于网络药理学筛选出了麻杏化瘀汤治疗儿童MPP的关键作用靶点和信号通路,这些靶点、通路与儿童MPP密切相关,各个通路、靶点之间相互联系、影响,产生协同效应,从侧面证实了麻杏化瘀汤治疗儿童MPP的复杂机制。 Objective To explore the possible molecular mechanism of main components of Maxing Huayu Tang(麻杏化瘀汤)in the treatment of mycoplasma pneumoniae pneumonia in children by means of network pharmacology.Methods The active constituents and related targets of Maxing Huayu Tang were retrieved and extracted from systematic pharmacology analysis platform of Traditional Chinese Medicine(TCMSP).The genes related to mycoplasma pneumoniae pneumonia in children were screened out from the disease target screening databases Genecards and OMIM,and the potential targets for the treatment of Mycoplasma pneumoniae pneumonia by Maxing Huayu Tang were selected by intersecting with drug targets,and the drug-chemical compotion-key target network map was constructed.The protein interaction(PPI)network was mapped using STRING database and Cytoscape software.GO enrichment analysis and KEGG signaling pathway enrichment analysis were performed for key targets.Results A total of 126 active ingredients of Maxing Huayu Tang were selected,and 78 intersections of drug targets and disease targets were identified.GO enrichment yielded 632 items,including 497 biological processes,91 molecular functions and 44 cellular components.KEGG enrichment analysis showed 134 related pathways,which were mainly related to PI3K/Akt signaling pathway,MAPK signaling pathway,IL-17,TNF signaling pathway,and Th17 cell differentiation.Conclusion This study preliminary confirms that Maxing Huayu Tang can intervene in the treatment of mycoplasma pneumoniae pneumonia in children from multi-component,multi-target and multi-pathway,which provides the basis for subsequent research.
