人参皂苷对过氧化氢诱导的Caco-2细胞氧化损伤的保护作用

摘要 为研究人参皂苷对H_(2)O_(2)诱导的人结肠癌(Caco-2)细胞氧化应激损伤的保护作用,构建H_(2)O_(2)诱导的Caco-2细胞氧化损伤模型,以不同质量浓度(10、20、50μg/mL)的人参皂苷干预H_(2)O_(2)诱导后的Caco-2细胞,通过形态学观察,CCK-8法检测细胞活力,测定胞内超氧化物歧化酶(superoxide dismutase,SOD)和谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)水平,测定氧化损伤标志物丙二醛(malondialdehyde,MDA)活力以及活性氧(reactive oxygen species,ROS)水平,评价人参皂苷的抗氧化应激水平。结果表明,200μmol/L的H_(2)O_(2)诱导Caco-2细胞4 h后,细胞存活率达到58%左右,符合建模要求。添加10~50μg/mL的人参皂苷明显提升受损细胞的存活率、SOD以及胞内GSH-Px活性,降低受损细胞内ROS和MDA水平,对H_(2)O_(2)诱导的Caco-2细胞氧化应激损伤起到良好的保护作用。 This study aimed to study the protective effect of ginsenosides on oxidative stress injury of human colon cancer(Caco-2)cells induced by hydrogen peroxide.Human colon cancer Caco-2 cells were cultured to construct the oxidative damage model of Caco-2 cells induced by hydrogen peroxide.The oxidative stress model of Caco-2 cells treated by ginsenosides at different concentrations(10,20,and 50μg/mL)was used to observe the cell vitality by morphological observation and the CCK-8 method.The intracellular superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)levels were determined.The activity of oxidative damage marker malondialdehyde(MDA)and reactive oxygen species(ROS)were measured to evaluate the antioxidant stress level of ginsenosides.Results showed that the survival rate of Caco-2 cells reached about 58%after inducing Caco-2 cells with 200μmol/L H_(2)O_(2) for 4 h,which met the modelling requirements.Ginsenosides(10-50μg/mL)significantly increased the survival rate,SOD,and intracellular GSH-Px activity,and decreased the level of ROS and MDA in the damaged cells.Ginsenosides can protect Caco-2 cells from oxidative stress induced by hydrogen peroxide.
