摘要
目的 探讨润肺益肾饮对肺癌荷瘤大鼠的抑瘤作用和肿瘤免疫微环境的影响。方法 将Lewis肺癌细胞接种于大鼠右季肋部皮下建立肺癌大鼠模型,随机分为模型组、润肺益肾饮低剂量(润肺益肾饮-L)组(3.5 g/kg)、润肺益肾饮中剂量(润肺益肾饮-M)组(7.0 g/kg)、润肺益肾饮高剂量(润肺益肾饮-H)组(14.0 g/kg)以及阳性药组(6 mg/kg顺铂),并以正常大鼠为对照组;干预结束后,计算各组大鼠肺指数、脾脏指数及抑瘤率;收集血液,酶联免疫吸附试验(ELISA)检测血清中转化生长因子(TGF)-β、白细胞介素(IL)-2、γ干扰素(IFN-γ)水平;流式细胞术检测外周血T淋巴细胞亚群;分离肺肿瘤组织,HE染色检测其病理学变化,Western blot检测肿瘤组织程序性死亡受体1(PD-L1)和B淋巴细胞瘤-2(Bcl-2)表达。结果 与对照组相比,模型组病理损伤严重,肺指数、TGF-β、CD8^(+)、PD-L1、Bcl-2表达水平升高,脾指数、IL-2、IFN-γ、CD3^(+)、CD4^(+)及CD4^(+)/CD8^(+)降低(P<0.05);与模型组相比,润肺益肾饮-L组、润肺益肾饮-M组、润肺益肾饮-H组病理损伤逐渐得到改善,肺指数、瘤质量、CD8^(+)、TGF-β、PD-L1、Bcl-2表达水平降低,脾指数、IL-2、IFN-γ、CD3^(+)、CD4^(+)及CD4^(+)/CD8^(+)升高(P<0.05);润肺益肾饮-H组各指标与阳性药组差异无统计学意义(P>0.05)。结论 润肺益肾饮可显著抑制肺癌荷瘤大鼠的肿瘤生长,改善肿瘤免疫微环境。
Objective To explore the anti-tumor effect of Runfei Yishen drink on lung cancer bearing rats and its impact on tumor immune microenvironment.Methods A lung cancer rat model was established by injecting Lewis lung cancer cells subcutaneously into right quarter rib of rats,and model rats were randomly divided into the model group,the low dose Runfei Yishen drink(Runfei Yishen drink-L)group(3.5 g/kg),the medium dose Runfei Yishen drink(Runfei Yishen drink-M)group(7.0 g/kg),the high dose Runfei Yishen drink(Runfei Yishen drink-H)group(14.0 g/kg)and the positive drug group(6 mg/kg cisplatin).Normal rats were used as the control group.After the intervention,lung index,spleen index and tumor inhibition rate of rats were calculated in each group.Blood samples were collected,and serum levels of transforming growth factor-β(TGF-β),interleukin(IL)-2 and interferon g(IFN-g)were detected by ELISA.T lymphocyte subsets in peripheral blood were detected by flow cytometry.Lung tumor tissue was isolated and its pathological changes were detected by HE staining.The expression levels of programmed death receptor 1(PD-L1)and B-lymphoblastoma-2(Bcl-2)were detected by Western blot assay.Results Compared with the control group,the pathological injury of the model group was serious,the lung index,TGF-β,CD8^(+),and the expression levels of PD-L1 and Bcl-2 were increased,and the spleen index,IL-2,IFN-γ,CD3^(+),CD4^(+)and CD4^(+)/CD8^(+)were decreased(P<0.05).Compared with the model group,the pathological damage of the Runfei Yishen drink-L group,the Runfei Yishen drink-M group and the Runfei Yishen drink-H group gradually improved,the lung index,tumor weight,CD8^(+),TGF-βand the expression levels of PD-L1 and Bcl-2 were decreased,and spleen index,IL-2,IFN-γ,CD3^(+),CD4^(+)and CD4^(+)/CD8^(+)were increased(P<0.05).There were no significant differences in above indexes between the Runfei Yishen drink-H group and the positive drug group(P>0.05).Conclusion Runfei Yishen drink can obviously inhibit tumor growth and improve the tumor immu
