乳苣不同部位异槲皮苷含量测定及其抗氧化活性研究

摘要 采用高效液相色谱(HPLC)法测定了乳苣不同部位的异槲皮苷含量;以抗坏血酸(VC)为阳性对照,采用DPPH法和FRAP法对乳苣不同部位提取物的抗氧化活性进行了初步探索。结果表明,乳苣样品在14 min内分离度良好,异槲皮苷浓度在2.00~10.00μg·mL^(-1)范围内与吸光度线性关系良好(R=0.9996),精密度(RSD=0.63%)、重复性(RSD=1.24%)、平均加标回收率(95.15%,RSD=1.72%,n=6)均符合要求,乳苣全草提取物溶液在室温下放置24 h稳定性良好(RSD=1.75%);采用HPLC法测得乳苣茎、叶、全草提取物中异槲皮苷含量分别为0.67 mg·g^(-1)、1.70 mg·g^(-1)、1.21 mg·g^(-1),根中未检测到异槲皮苷;乳苣不同部位提取物均具有一定的抗氧化活性,其对DPPH自由基的清除率和还原能力均随提取物浓度的增加逐渐升高,特别是乳苣叶提取物,当其浓度为0.4 mg·mL^(-1)时,其还原能力接近VC。 We determined the contents of isoquercitrin in different parts of Mulgedium tataricum L.by high performance liquid chromatography(HPLC).Moreover,we preliminarily explored the antioxidant activity of extracts from different parts of Mulgedium tataricum L.via DPPH method and FRAP method by using ascorbic acid(VC)as a positive control.The results show that the separation of Mulgedium tataricum L.sample is good within 14 min,the linear relationship between isoquercitrin content and absorbance is good in the range of 2.00-10.00μg·mL^(-1)(R=0.9996).The precision(RSD=0.63%),the repeatability(RSD=1.24%),and the average recovery(95.15%,RSD=1.72%,n=6)meet the requirements.The extract solution of Mulgedium tataricum L.whole herbs is stable at room temperature for 24 h(RSD=1.75%).The contents of isoquercitrin in stems,leaves,and whole herbs are 0.67 mg·g^(-1),1.70 mg·g^(-1),and 1.21 mg·g^(-1),respectively,but no isoquercitrin is detected in the roots by HPLC.The extracts from different parts of Mulgedium tataricum L.have certain antioxidant activity,and their scavenging rates towards DPPH free radicals and reducing abilities increase gradually with the increase of extract concentration,especially when the concentration of extract from Mulgedium tataricum L.leaves is 0.4 mg·mL^(-1),its reducing ability is equivalent to that of VC.
