摘要
目的探究藏药二十五味鬼臼丸对去势大鼠血清性激素及骨转换标志物的影响。方法将36只SD大鼠随机分为给药组、模型组及假手术组,给药组和模型组做卵巢摘除手术。术后两周开始灌胃给药,给药组予二十五味鬼臼丸混悬液(400mg/kg),模型组和假手术组予蒸馅水,每日1次,持续12周。用微计算机断层扫描技术(Micro-CT)检测右侧股骨近端结构、扫描电子显微镜(SEM)观测股骨微结构;以酶联免疫吸附试验(ELISA)检测大鼠血清骨吸收标志物抗酒石酸酸性磷酸酶5b(TRAP5b),血清骨形成标志物骨碱性磷酸酶BALP,血清E,、LH、FSH。结果Micro-CT和SEM显示,藏药二十五味鬼臼丸能显著改善去势骨质疏松症大鼠模型的股骨近端骨微结构和骨小梁密度;Pearson相关性分析显示,血清E_(2)水平与TRAP5b水平呈负相关,与BALP水平呈正相关;血清FSH、LH水平与BALP水平呈负相关。结论藏药二十五味鬼臼丸对去势大鼠血清性激素的影响:升高血清E,激发机体下丘脑-垂体-卵巢轴(HPO)上游的LH、FSH反馈调节;对骨转换标志物的影响:降低血清TRAP5b含量同时升高血清BALP含量,可改善骨代谢失衡,进而改善骨微结构。
Objective To explore the effects of Xizang medicine Ershiwuwei Guijiu Pill on serum sex hor-mones and bone turnover markers in the ovariectomized rats.Methods 36 SD rats were randomly divided into med-icine group,model group and sham group.Ovariectomy was performed in the medicine group and model group.Two weeks after the surgery,the medication was administered by gavage,with the medicine group receiving a suspen-sion of Ershiwuwei Guijiu Pill(400 mg/kg),and the model group and sham operation group receiving distilled water once a day for 12 weeks.Micro-CT was used to detect the structure of the right proximal femur;SEM was used to observe the microstructure of the femur;ELISA was used to detect serum bone resorption marker tartrate-resistant acid phosphatase 5 b(TRAP5 b),serum bone formation marker serum bone-specific alkaline Phosphatase(BALP),serum estradiol(E_(2)),luteinizing hormone(LH),follicle-stimulating hormone(FSH).Results The results of Micro-CT and SEM showed that Ershiwuwei Guijiu Pill could significantly improve the proximal femur bone microstructure and bone trabecular density in the model of ovariectomized osteoporosis rats.The pearson correlation analysis resalts showed that there is a negative correlation between E,levels and TRAP5 b levels,while serum E,levels are positively correlated with BALP.There is a negative correlation between serum FSH,LH levels and BALP levels.Conclusion The Xizang medicine Ershiwuwei Guijiu Pill could improve the level of serum E,in the ovariectomized rats and stimulate the feedback regulation of LH and FSH in the upstream of hypothalamic pituitary ovarian axis(HPO).In addition,it could also reduce the content of serum TRAP5 b and increase the content of serum BALP,which could improve bone metabolism imbalance and thereby improve bone microstructure。
