摘要
目的研究一株G4P[6]基因型A组轮状病毒(group A rotavirus,RVA)CZ079的全基因组特征及进化规律.方法利用逆转录-聚合酶链式反应(reverse transcription-polymerase chain reaction,RT-PCR)对CZ079的VP7、VP4、VP6、NSP1节段分别进行RT-PCR扩增,通过在线RVA自动分型工具对其分型,采用DNAstar5.1和Mega 11.0软件对各节段进行同源性及遗传进化分析.结果CZ079毒株基因型为G4-P[6]-I5-R 1-C1-M1-A8-N1-T1-E1-H1.VP6为I5型,NSP1为A8型.CZ079与LLR^(TM)、RotaTeq^(TM)和Rotarix^(TM)三种疫苗株在VP7的抗原表位中7-1b区域均存在氨基酸差异;VP4蛋白裂解后的VP8*抗原表位中8-1、8-2、8-3、8-4、5-1区域存在较大差异.结论2022年RVA毒株CZ079为罕见的G4P[6]-I5-A8型,推测该基因型RVA是由越南等东南亚地区传入中国,与中国RVA发生重配产生的新型重配毒株.
Objective To study the whole genome characteristics and evolutionary pattern of a genotype G4P[6]strain of group A rotavirus(RVA)CZ079 in China.Methods The genomic segments of CZ079,VP7,VP4,VP6 and NSP1 were amplified by reverse transcription polymerase chain reaction(RT-PCR),and the genotyping used online RVA automatic typing tool.DNAstar 5.1 and MEGA 11.0 were used in homology and genetic evolution analyses.Results The genotype of the CZ079 strain was G4-P[6]-I5-R1-C1-M1-A8-N1-T1-E1-H1.The typing by VP6 was type I5,and type A8 by NSP1.The amino acid differences were found between CZ079 and all three vaccine strains of LLR™,RotaTeq™and Rotarix™in the region 7-1b within the antigenic epitopes of VP7.After degradation of VP4,the 8-1,8-2,8-3,8-4 and 5-1 regions of VP8*antigenic epitopes showed large differences.Conclusions The RVA strain CZ079 detected in China in 2022 was a rare strain of genotype G4P[6]-I5-A8.It is hypothesized that this genotype of RVA was imported to China from Vietnam and other Southeast Asian regions,and formed a new reassortment strain by reassortment with RVA strains in China.
