摘要
【目的】鉴定和分析苦荞白粉病病原菌,并确定其在白粉菌系谱中的位置。【方法】通过孢子形态观察、致病性测定及核糖体基因内转录间隔区(internal transcribed spacer,ITS)和核糖体大亚基(ribosomal large subunit,LSU)序列系统发育树分析对苦荞白粉病病原菌进行鉴定,并进行多序列比对和聚类分析。【结果】苦荞白粉病病原菌鉴定为蓼白粉菌(Erysiphe polygoni);经ITS多序列比对和聚类分析发现该白粉菌(607 bp,MW494930.1)与甜荞上的白粉菌(E.polygoni,KP076437.1)相似性为100.00%,与大黄、酸模、蓼、竹节蓼、珊瑚藤等蓼科植物上的16个菌株ITS扩增片段相似性均高于98.83%;基于LSU序列扩增获得1条长度为910 bp的片段(OK490143),与酸模上的白粉菌(E.polygoni,MT361769.1)序列相似性为99.89%。【结论】本研究将侵染苦荞的白粉菌鉴定为子囊菌门(Ascomycota)白粉菌目(Erysiphales)白粉菌属(Erysiphe)蓼白粉菌(E.polygoni)。研究结果可为苦荞白粉病的防治提供依据。
[Purpose]To identify and analyze the pathogen of tartary buckwheat powdery mildew and determine its position in the powdery mildew pedigree.[Methods]The tartary buckwheat powdery mildew pathogen was identified based on spore morphology observation,pathogenicity test and phylogenetic tree analysis of internal transcribed spacer(ITS)and ribosomal large subunit(LSU)sequence,and multiple sequence alignment and cluster analysis of sequences were carried out.[Results]The pathogen of powdery mildew of tartary buckwheat was identified as Erysiphe polygoni.Through ITS multiple sequence alignment and cluster analysis,it was found that the powdery mildew(607 bp fragment,MW494930.1)was 100.00%similar to E.polygoni(KP076437.1)on common buckwheat.The similarity of that ITS amplification sequences with 16 strains collected from rhu-barb,sorrel,polygonum,ribbonbush and coral vines were also higher than 98.83%.A 910 bp fragment(OK490143)was obtained based on LSU sequence amplification,which showed 99.89%similarity with the strain of the E.polygoni(MT361769.1)on sorrel.[Conclusion]The tartary buckwheat powdery mildew is identified as E.polygoni,which belonged to Erysiphe,Erysiphales,Ascomycota.It provides a basis for the control of tartary buckwheat.
