摘要
目的 研究缬草酸对DACT2基因启动子甲基化的影响以及对乳腺癌细胞体外转移活性的影响。方法 将细胞人乳腺癌细胞MDA-MB-231分为对照组(NC)和缬草酸组(VA),MTT法检测细胞增殖能力,Transwell实验分析细胞侵袭能力,划痕实验分析细胞迁移能力,3D成球实验分析细胞生长能力,实时荧光定量PCR(q-RTPCR)检测细胞DACT2 mRNA表达水平,通过甲基化特异性PCR(Q-MSP)分析DACT2启动子甲基化水平,蛋白免疫印迹分析细胞DACT2蛋白表达情况。结果 与对照组相比,缬草酸处理能降低乳腺癌细胞的增殖、迁移、侵袭、3D成球和集落形成能力。与对照组相比,缬草酸处理后,乳腺癌细胞的DACT2基因启动子甲基化水平降低,DACT2 mRNA和蛋白表达水平升高。结论 缬草酸通过抑制DACT2基因启动子甲基化抑制乳腺癌细胞的恶性生物学过程。
Objective To study the effect of valerenic acid on the methylation of DACT2 gene promoter and the metastatic activity of breast cancer cells in vitro.Methods Cells were divided into control group(NC)and valerenic acid group(VA).The proliferation ability of valerenic acid on breast cancer cells was detected by MTT assay.The cell invasion ability was analyzed by Transwell assay.The cell migration ability was analyzed by scratch assay.The cell growth ability was analyzed by 3D spheroidization assay.The DACT2 mRNA expression was analyzed by real-time fluorescence quantitative PCR(q-RTPCR).The methylation level of DACT2 promoter was analyzed by methylation specific PCR(Q-MSP),and the DACT2 protein expression was analyzed by Western blot.Results Compared with the control group,valerenic acid treatment can reduce the proliferation,migration,invasion,3D spheroidization and colony formation ability of breast cancer cells.Compared with the control group,after valerenic acid treatment,the methylation level of DACT2 gene promoter in breast cancer cells decreased,and the expression levels of DACT2 mRNA and protein increased.Conclusion Valerenic acid can inhibit the malignant biological process of breast cancer cells by inhibiting the methylation of DACT2 gene promoter.
