摘要
[目的]建立藜麦二倍体苍白茎藜的组织培养体系。[方法]以二倍体苍白茎藜种子为外植体,先获得无菌幼苗,在此基础上进行不同脱毒外植体不同激素浓度配比对愈伤组织的诱导、不定芽分化的诱导、生根的诱导及移栽驯化的相关研究。[结果]二倍体苍白茎藜的种子最佳消毒时间为2%次氯酸钠消毒6 min;愈伤组织诱导的最佳培养基为MS+1.0 g/L水解酪蛋白+35 mg/L肌醇+690 mg/L脯氨酸+1.0 mg/L 6-BA+1.0 mg/L NAA+1.0 mg/L 2,4-D,诱导率达到86.63%;不定芽分化诱导培养基为MS+1.0 mg/L 6-BA+0.2 mg/L NAA+1.0 mg/L 2-IP+1.0 mg/L KT,诱导率为11.67%;MS+2.0 mg/L IBA或MS+0.5 mg/L IBA+0.1 mg/L NAA为最佳生根培养基,生根率均达100%,试管苗经炼苗和移栽驯化后成活。[结论]该研究通过苍白茎藜种子诱导获得再生植株,经炼苗移栽驯化后成活,为二倍体藜麦的遗传转化体系的建立提供基础。
[Objective]In order to establish the tissue culture system of diploid Chenopodium pallidicaule.[Method]Researches of callus induction,adventitious bud differentiation,rooting and transplanting were conducted by using the seeds of diploid Chenopodium pallidicaule.[Result]The results showed that the best disinfection time of diploid C.pallidicaule seeds was 6 min under 2%sodium hypochlorite;the callus induction medium was MS+1 g/L hydrolyzed casein+35 mg/L inositol+690 mg/L proline+1.0 mg/L 6-BA+1 mg/L NAA+1.0 mg/L 2,4-D,the induction rate reached 86.63%;the induction medium of adventitious bud differentiation was MS+2.0 mg/L 6-BA+0.2 mg/L NAA+1.0 mg/L 2-IP+1.0 mg/L KT,and the induction rate was 11.67%;MS+2.0 mg/L IBA or MS+0.5 mg/L IBA+0.1 mg/L NAA were the best rooting medium,with the rooting rate were 100%.[Conclusion]Regenerated plants were obtained from the explants of C.pallidicaule seeds,and then survived after being domesticated,which provided a basis for the establishment of the genetic transformation system of diploid C.pallidicaule.
