摘要
目的良种繁育能力的提升可从源头保障中药材质量。揭示影响丹参根段发芽的内在和外在环境因子,并对根段扩繁后种苗发育情况进行评价以确定根段扩繁最优条件。方法采用不同种质(A)(品系1、品系2、品系3)、根段直径(B)(<6mm、6~9mm、>9mm)、生根粉浓度(C)(0mg/g、10mg/g、20mg/g)、挖-切时间(D)(12月份挖-1月份剪切移栽、1月份挖-1月份剪切移栽、2月份挖-2月份剪切移栽)、种植深度(E)(3cm、6cm、9cm)、基质类型(F)(砂土、新营养土、旧营养土)和催芽场所(G)(泡沫箱、温室大棚、大田)共7个因素进行L18(3^(6)×6^(1))正交试验设计,探究影响丹参根段繁育的主要因素,并以地上茎长、幼芽重、生根数目和长茎数目4个指标评价幼苗的生长势。结果通过方差分析和多重比较(Duncan法),发现丹参根段发芽率与种质类型、根段直径、种植深度、基质类型和催芽场所间均存在显著相关性(P<0.05),与丹参幼苗的地上茎长、幼芽重、生根数目和长茎数目4个评价指标间均呈现显著正相关(P<0.01,R>0)。丹参种质差异和根段直径为影响根段发芽率的主要内部因素,催芽场所和生长环境的稳定性是保障丹参发芽和后续生长最重要的外在要素。为获得根段最高发芽率和最优生长势,除选育优良种质外,生产中可选择根段直径>9 mm、生根粉浓度10 mg/g、12月份采挖-1月份剪切、种植深度9 cm、以营养土为基质的扩繁方案。结论该研究首次开展了丹参扩繁效率及种苗生长的多因素评价,建立的丹参根段繁育优选方案可有效提高丹参根段发芽率并获得优良单株,适用于丹参工厂化和规模化种苗繁育。
Objective To determine the optimal conditions for root segment propagation by reveding the intrinsic and estrinsic factors affecting the germination of Salvia miltiorrhiza root segment,and evaluating the derelopment of seedlings after root segment propagation.Methods In this study,different Danshen strains(A)(strain 1,strain 2,strain 3),root segment diameters(B)(<6mm,6~9mm,>9mm),concentrations of root stimulants(C)(0mg/,10mg/g,20mg/g),digging-cutting time(D)(Dig in December-cut transplanting in January,dig in January-cut transplanting in January,dig in February-cut transplanting in February),burial depths(E)(3cm,6cm,9cm),soil types(F)(sandy soil,neotrophic soil,old trophic soil)and germination sites(G)(foam box,greenhouse,field)were used as seven factors for a multilevel orthogonal experiment,to explore the optimum conditions for germination of Salvia miltiorrhiza root segments.Meanwhile,the seedlings were evaluated by above-ground stem length,root number,young shoot weight,and long stem number.Results The germination rate was significantly correlated(P<0.05)with the factors of strain,root segment diameter,burial depth,soil type and germination site.There were significant positive correlations among the four evaluation indexes of above ground stem length,root number,young shoot weight,and long stem number of Salvia miltiorrhiza seedlings(P<0.01,R>0).Among them,strains and the root segment diameter,and nutrition storage of Salvia miltiorrhiza were the most important internal factors,whereas the germination site,was the most important external factor.Considering the influences on both the germination rate and seedling growth of Salvia miltiorrhiza,the favorite conditions were proposed in this study:propagating in foam boxes filled with nutrient soil,harvesting Salvia miltiorrhiza in December or January and cutting the primary lateral roots into segments of 9 mm in diameter in January,dipping into the solution of root stimulants at a concentration of 10 mg/g,and burying into the soil at a depth of 9 cm.Conclusion Th
