威灵仙栽培品和野生品的齐墩果酸含量测定及指纹图谱研究

Determination of the Oleanolic Acid Content and Establishment of Fingerprints of Cultivated and Wild Weilingxian(Clematidis Radix Et Rhizoma)

摘要 目的 考察人工栽培的威灵仙能否达到2020版《中华人民共和国药典》(一部)对齐墩果酸含量不得低于干燥品0.3%的要求,同时比较栽培品和野生品中齐墩果酸的含量差别。建立威灵仙的指纹图谱,分析栽培品和野生品的差异性。方法 参照2020版《中华人民共和国药典》(一部)测定样品的齐墩果酸含量,通过聚类分析和非配对样本t检验比较栽培品和野生品的含量差别。基于HPLC(DAD)建立威灵仙75%乙醇-水提取物的指纹图谱,通过相似度评价、主成分分析(PCA)、正交偏最小二乘法-判别分析(OPLS-DA)确定样品之间的差异性。结果 含量测定结果显示,栽培品能达到2020版《中华人民共和国药典》(一部)关于其含量的要求,且栽培品与野生品齐墩果酸含量差异无统计学意义(P>0.05)。剔除特殊样品后建立的指纹图谱共匹配出9个共有峰,通过相似度评价发现产自辽宁省西南部的样品和产自辽宁省东北部的样品化学成分差异较大。通过PCA和OPLS-DA发现建立指纹图谱的方法能够对栽培品和野生品进行区分,并确定出保留时间(RT)为13.984、55.772、70.646、84.891 min的峰为差异性色谱峰。差异性色谱峰峰面积比较结果显示栽培品RT为13.984 min的差异性色谱峰峰面积明显高于野生品(P<0.05),其余3个差异性色谱峰峰面积均明显低于野生品(P<0.05)。结论 研究发现从齐墩果酸含量的角度无法区分威灵仙的植物基源。建立的指纹图谱能够方便快速地区分威灵仙的植物基源,为威灵仙的质控和高效利用提供参考。 Objective To examine whether the artificially cultivated Weilingxian(Clematidis Radix Et Rhizoma)can meet the requirements of the 2020 edition of Pharmacopoeia of the People's Republic of China(Part I)that the content of oleanolic acid should not be less than 0.3%of the dried product,and to compare the difference in the content of oleanolic acid between cultivated product and wild product.To establish the fingerprints of Weilingxian(Clematidis Radix Et Rhizoma)and analyze the differences between cultivated product and wild product.Methods The content of oleanolic acid in the samples was determined with reference to the 2020 edition of PharmacopoeiaofthePeople'sRepublicofChina(Part I),and the differences in the content of cultivated product and wild product were compared by cluster analysis and t-test of unpaired samples.The fingerprints of the 75%ethanol-water extracts of Weilingxian(Clematidis Radix Et Rhizoma)were established based on HPLC(DAD),and the differences between the samples were determined by similarity evaluation,principal component analysis(PCA)and orthogonal partial least squares-discriminant analysis(OPLS-DA).Results The results of the content determination showed that the cultivated product could meet the requirements of the 2020 edition of Pharmacopoeia of the People's Republic of China(Part I)regarding its content,and there was no significant difference in the content of oleanolic acid between the cultivated product and the wild product(P>0.05).The fingerprints established after excluding the special samples were matched with 9 common peaks,and the similarity evaluation revealed that the samples from southwestern Liaoning province and those from northeastern Liaoning province had large differences in chemical composition.Through PCA and OPLS-DA,it was found that the method of establishing fingerprints in this study could distinguish the cultivated product and wild product,and the peaks with retention times of 13.984 min,55.772 min,70.646 min,and 84.891 min were identified as differential chroma
