摘要
目的研究苦豆子总碱的急性毒性,探讨苦豆子总碱对大鼠学习记忆功能和乙酰胆碱(Ach)生成的影响。方法采用急性毒性试验法测定苦豆子总碱对小鼠的急性毒性。将48只SPF级♂SD大鼠随机均分为正常对照组、苦豆子总碱组(低、中、高剂量分别为4、8、16 mg·kg^(-1)),连续灌胃给药28 d。观察大鼠的体重和体温变化;采用避暗试验检测大鼠的学习记忆功能;采用比色法检测血清与脑组织中Ach、乙酰胆碱酯酶(AChE)及胆碱乙酰移位酶(ChAT)的含量或活性。结果苦豆子总碱的小鼠急性经口半数致死量为0.203±0.024 g·kg^(-1)。与对照组比较,苦豆子总碱能明显降低大鼠的体温,延长大鼠的避暗潜伏期和减少大鼠的避暗次数,同时,降低大鼠血清和脑组织中Ach的含量和ChAT的活性及升高大鼠脑组织中AChE的活性,且降低脑组织中ChAT/AChE的比值。结论苦豆子总碱具有明显的急性毒性反应,对大鼠学习记忆功能有损伤,这可能与苦豆子总碱不仅降低大鼠体温,而且降低ChAT的活性,提高AChE的活性,从而导致大鼠的学习记忆功能受损,ACh的含量下降。
OBJECTIVE To study the acute toxicity and the effects of total alkaloids of Sophora alopecuroides on learning and memory function and on acetylcholine production in rats.METHODS The acute toxicity of total alkaloids of S.alopecuroides was determined by median lethal dose method in mice.Forty-eight male SD rats were randomly divided into normal control group,4 mg·kg^(-1)total alkaloids of S.alopecuroides group,8 mg·kg^(-1)total alkaloids of Sophora alopecuroides group,and 16 mg·kg^(-1)total alkaloids of S.alopecuroides group,with 12 animals in each group,were given continuous intragastric administration for 28 days.The weight and the body temperature of rats were observed.Dark-avoidance test was used to detect the learning and memory function of rats.Colorimetric method was used to detect Acetylcholine(ACh)content,acetylcholinesterase(AChE)activity and choline acetyltransferase(ChAT)activity in serum and brain tissues.RESULTS The results showed that the acute oral LD_(50)of total alkaloids of S.alopecuroides in mice was 0.203±0.024 g·kg^(-1).Compared with the normal control group,the total alkaloids of S.alopecuroides significantly decreased the body temperature of rats.They prolonged the dark-avoidance latency of rats and reduced the number of dark-avoiding times in rats.At the same time,the total alkaloids of S.alopecuroides reduced ACh content in the serum and brain of rats,significantly decreased ChAT activity,increased AChE activity and decreased the ChAT/AChE ratio in brain tissues.CONCLUSION The total alkaloids of S.alopecuroides have obvious acute toxic reactions and have a damaging effect on the learning and memory function in rats.This may be related to the fact that the total alkaloids of S.alopecuroides not only reduce the body temperature of rats,but also reduce ChAT activity and increase AChE activity,leading to impaired learning and memory in rats and a decrease in ACh content.
