摘要
目的 探讨叉头框O组转录因子3(FOXO3a)基因遗传多态性与某地区白癜风易感风险的相关性。方法 将2020年3月—2023年2月收治的197例白癜风患者纳入白癜风组,另将同期250例健康人纳入健康组。采用限制性片段长度多态性聚合酶链反应技术鉴定FOXO3a基因rs4946936位点基因分型;采用Hardy-Weinberg遗传平衡度检验评估样本代表性;白癜风发生的危险因素采用多因素Logistic回归分析。结果 2组TT、CT、CC基因型均符合Hardy-Weinberg遗传平衡定律(P>0.05)。2组TT、CT、CC基因型分布频率比较差异有统计学意义(P<0.01)。2组T、C等位基因分布频率比较差异有统计学意义(P<0.01)。多因素Logistic回归分析显示,皮肤外伤史、白癜风家族史、FOXO3a基因rs4946936位点TT基因型、FOXO3a基因rs4946936位点T等位基因是白癜风发生的危险因素(P<0.01)。结论 FOXO3a基因rs4946936位点多态性与白癜风易感风险密切相关,且FOXO3a基因rs4946936位点TT基因型、T等位基因是白癜风易感风险增加的危险因素。
Objective To investigate the correlation between the genetic polymorphism of FOXO3a gene and the susceptibility to vitiligo in a certain area.Methods A total of 197 patients with vitiligo admitted to our hospital from March 2020 to February 2023 were included in the vitiligo group,and 250 healthy people who underwent health examination during the same period were included in the health group.rs4946936 genotype of FOXO3a gene was identified by PCR with restriction fragment length polymorphism.Hardy-Weinberg genetic balance test was used to evaluate sample representativeness.The risk factors of vitiligo were analyzed by Logistic regression.Results The TT,CT and CC genotypes of the two groups were consistent with Hardy-Weinberg genetic balance law(P>0.05).There were significant differences in the distribution frequency of TT,CT and CC genotypes between the two groups(P<0.01).There were significant differences in the frequencies of T and C alleles between the two groups(P<0.01).Multiple Logistic regression analysis showed that skin trauma history,family history of vitiligo,TT genotype at rs4946936 site of FOXO3a gene,and T allele at rs4946936 site of FOXO3a gene were risk factors for incidence of vitiligo(P<0.01).Conclusion rs4946936 polymorphism of FOXO3a gene is closely related to susceptibility risk of vitiligo,and TT genotype and T allele of rs4946936 of FOXO3a gene are risk factors for increased susceptibility to vitiligo.
